摘要
采用乙醇浸提法从新鲜桑叶中提取1-脱氧野尻霉素,利用正交试验得到最佳提取条件。对桑叶1-脱氧野尻霉素的进行初步纯化,并研究了其生物特性。结果表明,最佳乙醇提取工艺为乙醇体积分数65%,pH 3.5,料液比1∶11,提取温度55℃。在284nm处测得1-脱氧野尻霉素质量浓度为0.028g/L,1-脱氧野尻霉素的得率为0.065%,对蔗糖酶的抑制率为19.08%。经石油醚、正丁醇纯化后,桑叶1-脱氧野尻霉素质量分数为21.84%,得率为0.057%,对蔗糖酶的抑制率提升至38.48%。1-脱氧野尻霉素对蔗糖酶的抑制作用表现为非竞争性抑制,增加浓度对蔗糖酶的抑制作用也增强。
1-deoxynojirimycin was extracted from fresh mulberry leaves via the method of ethanol extraction.The influence factors were determined and the optimum extraction process was obtained by orthogonal test.1-deoxynojirimycin was preliminarily purificated,and its biological characteristics were studied.The results showed that the optimum conditions of extracting were ethanol volume fraction of 65%,pH 3.5,solid-liquid ratio of 1∶11,and extraction temperature of 55 ℃.The mass concentration and yield of 1-deoxynojirimycin were 0.028 g/L and 0.065%,respectively at 284 nm.The inhibition rate of sucrase was 19.08%.The mass fraction and yield of 1-deoxynojirimycin purified by petroleum ether and n-butyl alcohol were 21.84%and 0.057%,while the inhibition rate of sucrase could reach to 38.48%. The effect of 1-deoxynojirimycin on sucrase showed noncompetitive inhibition,which was enhanced with the increasing concentration.
