摘要 目的探讨血液和尿液不同类型标本中巨细胞病毒(CMV)-DNA的检测研究。方法采用实时荧光定量聚合酶链反应(FQ-PCR)对52例患者4种不同类型的血液标本和85例患者3种不同类型的尿液标本的CMV-DNA载量进行检测,比较血液和尿液不同类型标本的CMV-DNA检出率和病毒载量的差异。结果 52例患者血清、全血、血浆和外周血单个核细胞(PBMC)的CMV-DNA检出率分别为48.08%、71.15%、57.69%和69.23%;全血和PBMC的CMV-DNA检出率较高,差异有统计学意义(P<0.05)。PBMC的定量结果较高。85例患者混合尿、尿上清和尿沉渣的CMV-DNA检出率分别为72.94%、62.35%和84.71%,尿沉渣的CMV-DNA检出率较高,混合尿的定量结果较高。结论血液和尿液标本CMV-DNA的检测结果差异较大,在CMV感染的临床诊疗和监测中使用同一标本类型检测具有重要的临床意义。 Objective To study the detection of cytomegalovirus(CMV)-DNA in different kinds of blood and urine sample.Methods The CMV-DNA loads in 3 different kinds of blood sample from 52 patients and 3 different kinds of urine sample from 85 patients were detected by real-time fluorescence quantitative polymerase chain reaction(FQ-PCR).The differences in CMV-DNA detection rate and virus loads were compared among different kinds of blood and urine samples.Results The CMV-DNA detection rates in serum,whole blood,plasma and peripheral blood mononuclear cell(PBMC)from 52 patients were 48.08%,71.15%,57.69% and69.23% respectively.The CMV-DNA detection rates of whole blood and PBMC were higher,the difference was statistically significant(P<0.05).The quantitative results of PBMC was higher.The CMV-DNA detection rates of mixed urine,urine supernatant and urine sediment from 85 patients were 72.94%,62.35% and84.71%respectively,the CMV-DNA detection rate of urine sediment was higher,while the quantitative results of mixed urine was higher.Conclusion The CMV-DNA detection results of blood and urine have large difference,therefore using the same kind of sample for conducting detection has an important clinical significance in the clinical diagnosis,treatment and monitoring of CMV infection.
