摘要
目的:研究断体三天的地龙提取液(DL3)对小鼠胚胎成纤维细胞(NIH3T3)增殖作用的影响。方法:制备断体三天的地龙提取液,盐析后得到四个组分(简称为S1、S2、S3、S4),以给药后成纤维细胞的增殖率为筛选指标,采用CCK-8试剂盒测定成纤维细胞的增殖率,观察不同组分对成纤维细胞增殖作用的影响,筛选出活性最强的组分。结果:在一定范围内,细胞增殖率随着蛋白浓度的增加呈依赖上升趋势。当蛋白浓度在6~8μg/m L之间时,各组间细胞增殖率均达到最大值,分别为113.62%、108.58%、117.03%和111.26%,其中S3对成纤维细胞增殖的促进作用最强。结论:断体再生期的地龙含有促进成纤维细胞增殖的活性组分,为进一步研究断体地龙中促进创伤愈合的物质奠定基础。
Objective: To investigate the proliferative effect of the extracts from tail-amputated earthworm for three days( DL3) on embryonic fibroblast( NIH3 T3). Methods: DL3 was separated into four fractions( S1,S2,S3,S4) by salting-out. The proliferation rates of NIH3 T3 were detected by Cell Counting Kit-8( CCK-8) to screen out the active components. Results: It showed that the cell proliferation rates were on the rise with the increasing protein concentration in a certain range. The proliferation rates of four groups reached the peak( 113. 62%,108. 58%,117. 03%,and 111. 26%) at the concentration of 6 ~ 8 μg/m L,and S3 was the most active fraction. Conclusion: DL3 is effective in promoting proliferation of NIH3 T3 cell,which lays the foundation for the further research on wound healing.
