香菇灵仙1号菌株提纯复壮 被引量:1

摘要 以发生退化的香菇(Lentinula edodes)灵仙1号为出发菌株,采用含有结晶紫的培养基(20%马铃薯,30mg/L结晶紫,2%葡萄糖,1%木屑粉,0.3%磷酸二氢钾,0.2%蛋白胨,0.15%硫酸镁,1.2%琼脂,pH自然)结合尖端菌丝分离方法进行提纯复壮。获得5个提纯复壮菌株,其中菌株7-2,与出发菌株相比,子实体产量提高12.6%,出菇提前15d,并恢复了灵仙1号正常状态下的"矮坛子"状——粗柄硬质菇特征。较出发菌株,5个提纯复壮菌株的木质素和纤维素降解能力均提高。 Hyphal tip isolation on a culture medium containing 20% potato, 30 mg/L crystal violet, 2% glucose, 1% wood powder, 0.3% KH2 PO4, 0.15% MgSO4, 0.2 % peptone and 1.2% agar (pH unadjusted) was used to obtain five rejuvenated strains of the Lentinula edodes cultivar, Lingxian 1, which had undergone degeneration. One rejuvenated isolate, strain 7-2 exhibited a 12.6% increase in fruit body yield, a 15-day reduction in the time from inoculation to fruiting, and an improvement in fruit body morphology (thicker stipe and harder fruit body) compared with the degenerated parent strain. All of the rejuvenated isolates also exhibited small increases in lignin-and cellulose-degrading capacities.
