摘要
研究从皱皮木瓜中提取的水溶性多糖的分离、纯化、单糖组成及结构,为木瓜的开发利用提供依据.采用Sevag法除蛋白、透析法除小分子杂质后,经Sephadex G-100葡聚糖凝胶柱分离得到了皱皮木瓜多糖CSP1.通过高效凝胶渗透色谱、气相色谱、紫外光谱、红外光谱和核磁共振氢谱等分析手段初步确定了其组成和结构.皱皮木瓜多糖CSP1为均一组分,重均相对分子质量为27 622,由鼠李糖、木糖、葡萄糖、半乳糖4种单糖组成,摩尔比为0.042∶0.067∶0.860∶0.031,葡萄糖是主要成分,木瓜多糖CSP1结构中存在α-糖苷键.
The study on the isolation,purification,monosaccharides composition and structure of watersoluble polysaccharidesfrom Chaenomeles Specious ( Sweet. )Nakai will provide experimental foundation for the further development of chaenomeles. The refined polysaccharides CSP1 were obtained by Sevag method for deprotein,dialysis and Sephadex G100 gel column chromatography.HPGPC,GC,UV,IR and 1HNMR were used to investigate the constituents and structrue of CSP1. Results CSP1 was homogeneousand the relative molecular weight was 27 622. The monosaccharides were Rha,Xyl,Glu and Gal,respectively,in molar ratio of 0. 042∶ 0. 067∶ 0. 860∶ 0. 031. There exists αglucosidal bond in the polysaccharides,in which Glu is the main constituent.
