新罗丹明酰胺基硫脲荧光探针在水溶液中对Hg^(2+)的识别 被引量:2

摘要 设计并合成了一类罗丹明酰胺基硫脲Hg^(2+)荧光探针,通过~1HNMR谱和^(13)C谱进行结构表征。采用紫外-可见光谱和荧光光谱研究了探针RFS对Hg^(2+)的光谱响应。结果表明,探针罗丹明B酰胺基邻氟苯基硫脲(RFS)在水溶液中(HEPES缓冲溶液pH7.2)对Hg^(2+)表现出高选择性,并具有快速、灵敏的显色和Off-On荧光增强效应。探针与Hg^(2+)作用后,溶液颜色由无色变为红色,在1~10μmol/L范围内,探针的荧光强度与Hg^(2+)浓度呈线性关系。比较了因取代基的电子效应不同对探针灵敏度的影响,通过激光共聚焦成像实现了探针RFS在Hela细胞内对Hg^(2+)的识别检测,表明探针RFS可用于检测细胞内的汞离子。 Three of rhodamine-base fluorescent probes, which were confirmed by 1HNMR and 13 CNMR detection were designed and synthesized. The interactions between Hg2+ and probes were investigated via UV absorption spectroscopy and fluorescence spectrum. The results showed that the probe RFS has good selectivity, rapid, sensitive and off-on fluorescence responses to Hg2+ in water. After the addition of Hg2+ , the probes solution changed from colorless to red and showed a good linear relationship with Hg2+ in the range of 1 - 10 μmol/L. Additionally, the probes have the detection limit of 0. 76 × 10 -7, 0. 6× 10-7 and 1.7 × 10-7 mol/L, respectively, suggesting their high sensitivities for Hg2+ detection and showed that the different substituent' s electonic effect had influence on the sensitivity of the three probes. The probe RFS was successfully used to detect Hg2+ in Hela cells in aqueous solution by laser confocal scanning microscope.
