摘要
目的建立一种适合4种苯二氮类药物(硝西泮、艾司唑仑、阿普唑仑和地西泮)的提取分离和定性定量方法。方法用固相萃取技术处理含药血浆,色谱柱为月旭C18柱(4.6mm×200mm,5μm);流动相为甲醇-0.025mol·L-1乙酸铵(62∶38);流速为0.8m L·min-1;检测波长为230nm;柱温为室温。结果四种组分提取完全,杂质去除彻底,分离良好,在10ng·m L-1~5000ng·m L-1范围内峰面积与浓度呈良好的线性(r>0.998),绝对回收率96%~103%,相对回收率95%~104%,日内日间RSD均<4%。结论该色谱条件可同时测定血浆中4种苯二氮类药物,具有快速、简便、准确、可行等特点,适用于此类药物中毒患者的快速定性和定量分析。
OBJECTIVE To estabolish a rapid and simple analysis method for the determination of four benzo- diazepines (Nitrazepam, Estazolam, Alprazolam, Diazepam) in human serum simuhaneously by HPLC coupled with UV detector after solid phase extraction. METHODS Four benzodiazepines in serum were extracted with SPE C18 column,separated on a YUEXU Cls column(4. 6mm ×200mm,51μm) at room temperature and eluted by mobile phase of methanol-0. 025mol· L-1 NH4AC (62:38) at flow rate of 0. 8mL· min-1. The detection wavelength was 230nm. Column temperature was room temperature. RESULTS The extraction was high in recovery and no interfer- ence existed. Four benzodiazepines were well separated under above chromatographic conditions. The excellent linear- ity of the four benzodiazepines was obtained with the concentration range of 10ng·mL-1- 5000ng · mL-1 ( r 〉 0. 9980). The absolute recoveries were 96% - 103%, and the relative recoveries were 95%- 104%. Intra-day RSD and inter-day RSD were all less than 4%. CONCLUSION This method is rapid, simple, accurate, sensitive and suitable for qualitaty and quantity the benzodiazepines in a purpose of therapy or prognosis of the abuse and suicide.
