摘要
目的建立鸡血藤Spatholobus suberectus的HPLC指纹图谱分析方法,为其质量控制提供依据。方法采用ZORBAX SB-C18柱(150 mm×4.6 mm,5μm),以乙腈(A)-0.2%磷酸水溶液(B)为流动相进行梯度洗脱,体积流量为1 mL/min,检测波长260 nm,柱温为30℃。并运用聚类分析和主成分分析对鸡血藤指纹图谱进行模式识别研究。结果建立了鸡血藤药材HPLC指纹图谱的共有模式,确定35个共有峰,并根据对照品指认6个共有峰。17批药材的相似度分析结果与聚类分析、主成分分析结果基本一致。结论 HPLC指纹图谱结合聚类分析、主成分分析可以快速、高效地评价不同产地鸡血藤药材的质量差异。
Objective To establish an HPLC method for the fingerprint analysis of Spatholobus suberectus,so as to provide evidence for the quality control of it.Methods Separation was performed on a ZORBAX SB-C(18) column(150 mm × 4.6 mm,5 μm) and the mobile phase was acetonitrile-0.2% phosphoric acid with gradient elution.The flow rate was 1 m/min and the wavelength was 260 nm.The column temperature was 30 ℃.The cluster analysis(CA) and principal component analysis(PCA) were applied to studying the HPLC fingerprint and chemical pattern recognition.Results The HPLC fingerprint of S.suberectus has been established with 35 common peaks,and six of common peaks were identified.The values of similarity evaluation mostly agreed with the result of CA and PCA.Conclusion The application of HPLC combined with CA and PCA could quickly and objectively evaluate the quality of S.suberectus from different regions.
