高效液相色谱法梯度洗脱法同时测定补血调经片中甘草素、美迪紫檀素、香附烯酮和α-香附酮含量 被引量:4

Simultaneous Determination of Liquiritigenin, Medicarpin, Cyperotundone and α-Cyperone in Buxue Tiaojing Tablets by HPLC Gradient Elution Method

摘要 目的建立高效液相色谱法(HPLC)梯度洗脱法同时测定补血调经片中甘草素、美迪紫檀素、香附烯酮和α-香附酮含量。方法采用Waters Symmetry C18色谱柱(250 mm×4.6 mm,5μm);流动相乙腈-0.1%磷酸溶液,梯度洗脱,体积流量1.0 ml/min;柱温30℃,检测波长分别为280 nm(甘草素、美迪紫檀素)和242 nm(香附烯酮、α-香附酮)。结果甘草素、美迪紫檀素、香附烯酮和α-香附酮分别在1.09~21.80、1.26~25.20、19.58~391.60、8.91~178.20μg/ml范围内线性关系良好(r≥0.9992),平均加样回收率分别为96.96%、98.88%、99.85%、99.53%,相对标准偏差(RSD)分别为1.03%、1.17%、0.83%和0.77%。结论HPLC梯度洗脱法准确可靠,可用于补血调经片的质量控制。 Objective To establish an HPLC gradient elution method for simultaneous determination of liquiritigenin,medicarpin,cyperotundone andα-cyperone in Buxue Tiaojing Tablets.Methods The chromatographic separation was carried out on a Waters Symmetry C18(250 mm×4.6 mm,5μm)column,with mobile phase composed of acetonitrile-0.1%phosphoric acid solution as a mobile phase in gradient elution.The flow rate was 1.0 mL·min-1,the column temperature was 30℃.The detection wavelength were 280 nm for liquiritigenin and medicarpin,and 242 nm for cyperotundone andα-cyperone.Results Liquiritigenin,medicarpin,cyperotundone andα-cyperone showed good linear relationships within the ranges of 1.09-21.80,1.26-25.20,19.58-391.60,8.91-178.20μg·mL-1(r≥0.9992),whose average recoveries were 96.96%,98.88%,99.85%,99.53%with the RSDs of 1.03%,1.17%,0.83%,0.77%,respectively.Conclusion HPLC Gradient elution method is accurate and reliable,which can be used for the quality control of Buxue Tiaojing Tablets.
