小白菊内酯对人宫颈癌Hela细胞株生长与迁移的影响及其相关分子机制的研究 被引量:2

摘要 目的探讨小白菊内酯对人宫颈癌Hela细胞株生长与迁移的影响及其相关分子机制,为小白菊内脂的进一步抗肿瘤药物研发提供实验基础。方法应用CCK-8检测细胞活性,HE染色观察细胞形态学变化,细胞划痕实验检测细胞迁移能力的变化;细胞免疫化学染色检测BCL-2和NF—κB基因在细胞内表达,Western—blot方法检测细胞迁移相关蛋白Twist—relatedprotein1(Twistl)表达的变化。结果小白菊内酯作用后的Hela细胞的生长受到抑制,并诱导细胞凋亡。肿瘤细胞的迁移能力受到明显的抑制,实验组和对照组差异显著(P〈0.05),但是24h和48h差别并不明显;BCL-2和NF—κB基因在细胞内表达明显下调,Twistl表达明显下调,和Oh比较区别显著(P〈0.05),但是24h和48h并无明显区别。结论小白菊内酯能够抑制Hela细胞株生长与迁移能力,可能和抑制BCL-2,NF—κB基因与Twistl表达有关。 Objective To investigate the effect and possible molecular mechanisms of parthenolide on the growth and migra- tion of human cervical cancer gela cell line. Methods CCK - 8 was used to detect cell activity; cell morphological changes were ob- served by HE staining, the changes of cell migration was detected by cell scratchassay ; BCL - 2 and NF - κB were detected by immu- nocytochemistry; the expression changes of cell migration protein Twist - related 1 ( Twistl ) was detected by Western - blot assat. Results The growth of Hela cells was inhibited and the apoptosis was induced. The migration ability of tumor cells was inhibited. There was significant difference between the experiment group and the control group (P 〈 0. 05). But there was no significant differ-ence between 24h and 48h. BCL - 2. Compared with Oh, the expression of Twistl were down - regulated in cells ( P 〈 0. 05 ). Conehlsion Parthenolide can inhibit the growth and migration of Hela cell line, which may related to inhibit BCL - 2, and NF - κB is reahed to Twist 1.
