煎煮时间对防风中升麻素苷和5-O-甲基维斯阿米醇苷含量的影响 被引量:2

摘要 目的:通过测定不同煎煮时间对防风药液中有效成分升麻素苷、5-O-甲基维斯阿米醇苷含量的影响,探讨防风的最佳煎煮时间,为中药特殊煎煮方法临床应用提供理论依据。方法:采用HPLC法测定。色谱条件:Inertsil ODS-3柱(250×4.6 mm,5μm);流动相为甲醇-水(40:60),流速为1 mL·min-1,检测波长为254nm,柱温30℃。结果:防风的有效成分升麻素苷浓度在0.1-0.8mg/mL、5-O-甲基维斯阿米醇苷浓度在0.1-0.5mg/m L范围内与峰面积呈良好的线性关系(r=0.9992,r=0.9997)。防风煎煮20min升麻素苷含量以及5-O-甲基维斯阿米醇苷含量为4.712mg/g和1.860mg/g,煎煮25min为7.316mg/g和3.472mg/g,煎煮30min为7.564mg/g和3.472mg/g,煎煮35min为5.828mg/g和3.472mg/g,煎煮40min为5.456mg/g和2.976mg/g,煎煮45min为5.332mg/g和2.356mg/g,煎煮50min为4.588mg/g和1.860mg/g,煎煮55min为3.844mg/g和1.736mg/g,煎煮60min为3.224mg/g和1.736mg/g。结论:防风饮片煎煮30min时,其有效成分升麻素苷的含量最高,在煎煮25~35min时5-O-甲基维斯阿米醇苷含量最高且基本稳定,表明煎煮30min时有效成分总溶出率最高,同时,如果煎煮时间过低,其有效成分不能充分溶出;而且,如果煎煮过程过长,同样会降低其主要成分的溶出率;故临床应用防风以先煎10min为宜。药喷雾技术,以便遏制西洋参病虫害,确保苗株健壮,促成西洋参种植业的优质、高产和高效。 Objective: By determining the effect of decoeting time on the content of Prim-o-glucosylcimifuginand 5-O-methylvisammioside in radix sileris, and to explore the optimum boiling time of radix sileris which provides theoretical basis for clinical application of special decoction method of traditional Chinese medi- cine. Methods:By HPLC. The chromatographic condition of Puerarin contains the Inertsil ODS-3 column (250×4.6 mm,5 μm), mobile phase: methanol-water (40:60),the flow rate of lml .min,the detection wavelength of 254nm and the column temperature of 30 ℃.Results:Prim-o- glucosylcimifugin's concentration was 0.1-0.8mg/mLand 5-O-methylvisammioside'sconcentration was 0.1-0.5mg/mLwithin the scope of concentration had good linear relationship with the peak area (r= 0.9992,r-=0.9997). radix sileris decoct 20min Prim-o-glucosyleimifuginand 5-O-methylvisammioside content for the4.712mg/g and 1.860rag/g, decoct 25min for the 7.316mg/g and 3.472mg/g, decoct 30min for the 7.564mg/g and 3.472mg/g, decoct 35min for the 5.828mg/g and 3.472mg/g, decoet 40min for the 5.456mg/g and 2.976mg/g, decoct 45min for the5.332mg/g and 2.356rng/g, decoct 50min for the4.588mg/g and 1.860mg/g, decoet 55min for the 3.844mg/g and 1.736mg/g, decoet 60rain for the 3.224mg/g and 1.736mg/g. Conclusion: When radix sileris deeoct 35 min, the content of Prim-oglucosyleimifugin is the highest, when radix sileris deeoct 25-35min,the content of 5-O- methylvisammioside is high and stabile,the decocting time is too short active ingredientcan not be fully dissolved or too long of theactive ingredient content decreased, Clinical application of radix sileris to First fried 10min is appropriate.
