摘要
目的探索桂芍子喘颗粒混合挥发油的β-环糊精最佳包合工艺,并对其稳定性进行研究。方法采用正交试验,以挥发油包合率为指标考察饱和水溶液法包合工艺;通过X射线衍射法、显微成像分析法对包合物进行表征;分别对包合物和物理混合物进行高温、高湿、挥发性实验,以挥发油相对含量和失重百分率为指标,考察其稳定性。结果挥发油包合工艺为挥发油与β-CD的配比为1∶10,40℃的包合温度,进行3 h包合;X射线衍射法和显微成像分析法均证明混合油包合物已经形成;在热、湿因素影响下,桂芍子喘颗粒混合挥发油β-环糊精包合物与物理混合物进行比较,前者的稳定性有显著提高。结论优化挑选的挥发油包合工艺是稳定可行的,可以用于桂芍子喘颗粒的生产。环糊精包合技术减少了颗粒制备中挥发油的损失,改善了挥发油的热稳定性和湿稳定性,适用于含挥发油的固体制剂生产。
Objective To optimize the prepration process of β-cyclodextrin(β-CD) inclusion compound in Guishao Zi Chuan Granules,and research its stability.Methods Orthogonal test is used to investigate the inclusion rate of volatile oil as an index;Both X-ray diffraction and microscopic imaging assays have shown that the inclusion compound has been formed;The high temperature,high humidity and volatile experiments are carried out on the inclusion compound and the physical mixture.The stability of the inclusion compound and the physical mixture are investigated by the relative content of volatile oil and the percentage of weight loss.Results The inclusion process is the inclusion temperature of the volatile oil and β-CD at a ratio of 1∶10 and 40℃ for 3 h.X-ray scattering technique and X-ray scattering technique preliminary proved the inclusion compound had been formed.Under the influence of heat and humidity factors,the stability of the β-cyclodextrin inclusion compound of the volatile oil is much better than that of the physical mixture.Conclusion It is stable and feasible to optimize the inclusion process of volatile oils,and the stable inclusion complex can be obtained.The inclusion technique is suitable for improving the thermo-stability,hygro-stability and anti-volatility of the mixed volatile oil.
