摘要
目的:了解注射液雷替曲塞对胃癌患者癌细胞的影响及作用机制。方法:将购入的84只裸鼠建立人胃癌细胞MGC-803皮下移植瘤模型,建模成功后随机分为3组,常规组给予腹腔注射生理盐水,雷替曲塞组给予腹腔注射雷替曲塞,5-Fu组给予腹腔注射5-Fu,详细观察对比干预后裸鼠体重、移植瘤体积、瘤重、抑瘤率、凋亡率及癌细胞的周期分布情况。结果:常规组裸鼠体重、移植瘤体积、瘤重均高于雷替曲塞组与5-Fu组,差异有统计学意义(P<0.05);雷替曲塞组裸鼠体重高于5-Fu组,差异无统计学意义(P>0.05),雷替曲塞组移植瘤体积、瘤重低于5-Fu组,差异有统计学意义(P<0.05);常规组裸鼠癌细胞S期分布高于雷替曲塞组与5-Fu组,差异有统计学意义(P<0.05);雷替曲塞组与5-Fu组S期分布,差异无统计学意义(P>0.05);雷替曲塞组与5-Fu组G0/G1,差异无统计学意义(P>0.05);雷替曲塞组与5-Fu组G0/G1期分布低于常规组,差异有统计学意义(P<0.05);三组G2/M期分布无明显差异,差异无统计学意义(P>0.05);常规组癌细胞抑瘤率、凋亡率明显低于雷替曲塞组与5-Fu组,差异有统计学意义(P<0.05);雷替曲塞组癌细胞抑瘤率、凋亡率显著高于5-Fu组,差异有统计学意义(P<0.05)。结论:胃癌患者给予雷替曲塞治疗与5-Fu抑瘤效果相似,可有效抑制人胃癌细胞MGC-803裸鼠移植瘤生长,并可诱导癌细胞凋亡。
Objective：To investigate the effect and mechanism of injection of rahitrexed on cancer cells in gastric cancer patients. Methods： 84 nude mice were purchased to establish the human gastric cancer cell MGC-803 subcutaneous tumormodel, after successful modeling ,micewere randomly divided into 3 groups, the conventional group received intraperitoneal injection of saline, the rahitrexed group were given intraperitoneal injection of raltitrexed, the 5-Fu group were given intraperitoneal injection of 5-Fu, nude miceweight, tumor volume, tumorweight ,inhibition rate,apoptosis rate, cycle distribution of cancer cellsafter intervention were observed.
