摘要
目的探讨雷替曲塞联合阿帕替尼对结肠癌细胞增殖的影响及其机制。方法体外培养人结肠癌细胞SW480(以下称SW480细胞),取传3代、对数生长期、生长状态良好的SW480细胞,随机分为对照组、雷替曲塞0.5μg/mL组、雷替曲塞1.0μg/mL组、雷替曲塞1.5μg/mL组、阿帕替尼组、联合干预组,雷替曲塞0.5μg/mL组、雷替曲塞1.0μg/mL组、雷替曲塞1.5μg/mL组分别加入含0.5、1.0、1.5μg/mL雷替曲塞的DMEM培养液,阿帕替尼组加入含1.5μg/mL阿帕替尼的DMEM培养液,联合干预组加入含1.5μg/mL雷替曲塞和1.5μg/mL阿帕替尼的DMEM培养液,对照组加入等量DMEM培养液。各组分别于37℃、5%CO 2、饱和湿度的细胞培养箱孵育48 h,收集细胞,采用MTT法检测细胞增殖活性,计算细胞增殖抑制率;采用实时荧光定量PCR法检测Bax、Caspase-3、Bcl-2 mRNA表达,采用Western blotting法检测Bax、Caspase-3、Bcl-2蛋白表达。结果随着雷替曲塞浓度升高,SW480细胞增殖活性逐渐降低,细胞增殖抑制率逐渐升高,以联合干预组效果更明显(P均<0.05)。随着雷替曲塞浓度升高,SW480细胞Bax、Caspase-3 mRNA和蛋白相对表达量逐渐升高,而Bcl-2 mRNA和蛋白相对表达量逐渐降低,以联合干预组Bax、Caspase-3、Bcl-2 mRNA和蛋白相对表达量变化更明显(P均<0.05)。结论雷替曲塞联合阿帕替尼能够抑制人结肠癌细胞增殖,其效果优于雷替曲塞或阿帕替尼单独用药,其作用机制可能与调控Bax、Caspase-3、Bcl-2表达,促进肿瘤细胞凋亡有关。
Objective To investigate the effect of raltetrexed combined with apatinib on the proliferation of colon cancer cells and its mechanism.Methods Human colon cancer cells SW480(hereinafter referred to as SW480 cells)were cultured in vitro.SW480 cells of the third generation in the logarithmic growth stage and in good condition were taken and randomly divided into the control group,0.5μg/mL raltetrexed group,1.0μg/mL raltetrexed group,1.5μg/mL raltetrexed group,apatinib group,and combined group.The cells in the 0.5μg/mL group,1.0μg/mL group and 1.5μg/mL group were added with DMEM medium containing 0.5,1.0 and 1.5μg/mL of raptetrexed,and the cells in the apatinib group were added with DMEM medium containing 1.5μg/mL of apatinib,respectively.DMEM medium containing 1.5μg/mL rattrexed and 1.5μg/mL apatinib was added to the combined group,and the same amount of DMEM medium was added to the control group.Each group was incubated in a cell incubator at 37℃,5%CO 2 and saturated humidity for 48 h,respectively.The cells were collected,and the proliferation activity was detected by MTT,and the proliferation inhibition rate was calculated.The mRNA expression levels of Bax,Caspase-3,and Bcl-2 were detected by real-time fluorescence quantitative PCR,and the protein expression levels of Bax,Caspase-3,and Bcl-2 were detected by Western blotting.Results With the increased concentrations of raltetrexed,the proliferation activity of SW480 cells decreased gradually,the inhibition rate of cell proliferation increased gradually,and the effect was the most obvious in the combined group(all P<0.05).The mRNA and protein relative expression levels of Bax and Caspase-3 in SW480 cells increased gradually with the increased concentrations of raltetrexed,while the mRNA and protein relative expression levels of Bcl-2 decreased gradually.The changes in the mRNA and protein relative expression levels of Bax,Caspase-3 and Bcl-2 of the combined group were the most significant(all P<0.05).Conclusion Raltetrexed combined with apatinib can in
