摘要
α硫辛酸是一种具有抗氧化作用的二硫醇活性化合物,能够保护骨细胞、成骨细胞,抑制破骨细胞生成和活性;同时,α硫辛酸可增加骨密度,提高骨质量。α硫辛酸抗骨质疏松的作用与抑制氧化应激、干扰NF-κB活化、下调RANKL和激活Wnt信号通路等有关。细胞和动物实验显示α硫辛酸能够促进骨形成和抑制骨吸收,具有治疗骨质疏松的作用,但是有关的临床研究很少,α硫辛酸最佳治疗剂量、疗程以及与其他抗骨质疏松药物联合治疗是否具有累加效应,需要进一步的研究证实。
Alpha lipoic acid is a dithiol bioactive compound with antioxidant properties. In vitro,alpha lipoic acid can protect osteocytes and osteoblasts,while inhibit osteoclastogenesis and osteoclast activity. Animal experiments and clinic studies found that alpha lipoic acid could increase bone mineral density and quality. Current data indicate that the anti-osteoporosis mechanism of alpha lipoic acid involves the inhibition of oxidative stress,interference with NF-κB activation,downregulation of RANKL and activation of Wnt signal pathway. Cellular and animal studies showed that alpha lipoic acid could promote bone formation and inhibit bone resorption,and could play a role in treating osteoporosis. However,clinical studies on alpha lipoic acid are limited,and the optimal dose,treatment duration,and additive effect with other osteoporosis treatment medications deserve further study.
