摘要
采用木瓜蛋白酶酶解法制备多棘海盘车多肽,并对多肽分级纯化,获得不同分子量分布范围的样品Pe1、Pe3、Pe5、Pe8、Pr14。选取金黄色葡萄球菌、枯草芽孢杆菌、白色念珠菌、大肠杆菌4种供试菌种采用滤纸片扩散法测定5种样品的抑菌活性,采用微量琼脂稀释法测定样品对每种供试菌种的最小抑菌浓度(MIC)。结果表明:Pe1、Pe3、Pe5、Pe8、Pr14对供试菌种均具有抑制作用,Pr14对金黄色葡萄球菌的抑菌活性表现最佳,最低抑菌浓度为0.156 mg/m L;Pe1对枯草芽孢杆菌和白色念珠菌抑菌活性表现最佳,最低抑菌浓度分别为0.312,0.625 mg/m L;Pe8对大肠杆菌抑菌活性表现最佳,最低抑菌浓度为0.625 mg/m L。
Polypeptides from Asterias amurensis were prepared by papain enzyme hydrolysis method,and different molecular weight distributions samples of Pe1,Pe3,Pe5,Pe8,Pr14 were obtained by classification and purification. Used Staphylococcus aureus,Escherichia coli,Bacillus subtilis and Candida albicans as the indicator bacteria,filter paper dispersion methods were used to determine antimicrobial activity,the minimum inhibitory concentrations( MIC) of samples were determined by agar dilution method. The results showed that all samples had inhibitory effects on testing bacteria,but the antimicrobial activity of different polypeptide on testing bacteria are different. The antimicrobial activity of the Pr14 sample on Staphylococcus aureus was the highest,and the MIC against Staphylococcus aureus was 0. 156 mg/m L; the antimicrobial activity of the Pe1 sample on Sacillus subtilis and Candida albicans was the highest,and the MIC were 0. 312 mg/m L and 0. 625 mg/m L; the antimicrobial activity of the Pe8 sample on Escherichia coli was the highest,and the MIC was 0. 625 mg/m L.
