摘要 目的:观察并探讨胰岛素对妊娠期糖尿病孕妇胎盘组织钙黏蛋白(VE-cadherin)、β-粘环素(β-catenin)、膜蛋白A2(Annexin A2)、膜蛋白A5(Annexin A5)、膜蛋白14-3-4(Annexin 14-3-4)表达的影响。方法:选择纳入妊娠期糖尿病患者(GDM组)和正常糖代谢孕妇(对照组)各10例为研究对象,采用免疫印迹(western blot)和免疫组化技术检测其胎盘组织VE-cadherin、β-catenin、Annexin A2、Annexin A5、Annexin 14-3-4蛋白的表达。结果:western blot结果表明,GMD组胎盘组织的Annexin A2、Annexin A5、VE-cadherin、Annexin 14-3-4表达较对照组明显下调(P<0.05)。免疫组化结果显示,GDM胰岛素治疗组Annexin A2、Annexin A5、VE-cadherin、Annexin 14-3-4较单纯饮食控制组表达高(均为P<0.05)。结论:饮食和胰岛素干预可以提高胎盘VE-cadherin、Annexin A2、Annexin A5和Annexin 14-3-4的表达,为GDM发病机制和治疗提供了理论依据。 Objective : To observe and investigate the effects of insulin on the expression of VE - cadherin, β - catenin, Annexin A2, Annexin AS, and Annexin 14-3 -4. Methods: 10 cases of gestational diabetes mellitus (GDM group) and normal glucose metabolism pregnant women ( control group) were treated with western blot and immunohistochemical technique to detect the expression of VE - cadherin, β - catenin, Annexin A2, Annexin A5, Annexin 14 -3 -4 protein expression. Results :The expression of Annexin A2, Annexin A5, VE -cadherin and Annexin 14 - 3 - 4 in placental tissue of GMD group was significantly lower than that of the control group ( P 〈 0.05 ). The expression of Annexin A2, Annexin AS, VE -cadherin and Annexin 14 -3 -4 in GDM insulin treatment group was higher than those in control group ( P 〈 0.05 ). Conclusion:Di- etary and insulin intervention can improve the expression of VE -cadherin, Annexin A2, Annexin A5 and Annexin 14 -3 -4 in placenta, and pro- vide the theoretical basis for the pathogenesis and treatment of GDM.
