摘要
研究利用荧光光度法测定葡萄皮水提取物抗氧化活性的新体系。在稀硫酸介质中,荧光染料吖啶红与Fenton反应产生的羟自由基反应后,产生荧光猝灭,利用葡萄皮水提取物对羟自由基的清除作用,使吖啶红荧光强度减弱,据此建立测定葡萄皮抗氧化能力的新方法,探讨了最佳实验条件,测定了不同品种葡萄皮水提取物的抗氧化活性。结果表明,玫瑰香葡萄皮水提取物的抗氧化能力最强,该法操作简便、测定快速、稳定性好,可作为判断天然提取物抗氧化能力的筛选方法。
A new system for determining the antioxidant activity of grape skin extract by simple fluorescence method was studied. The hydroxyl radical produced by Fenton reaction can decrease fluorescence intensity of acridine red in dilute sulphuric acid medium. As water extract of grape skin has scavenging effect on hydroxyl free radical, the acridine red fluorescence intensity can be weakened. According to this principle, a new method for the determination of grape skin's antioxidant activity was developed. The optimal experimental conditions were discussed, and antioxidant activities of different grape skin were determined. The results showed that the Muscat grape skin has the highest antioxidant activity. The method is simple, easy to operate, rapid to determine and has good stability, thus can be applied to determine antioxidant capacity of natural extract.
