摘要
目的探讨Sonic Hedgehog信号通路Ptch1和Ptch2(Ptch1/2)成员在鼻咽癌组织中的表达及其关系。方法应用免疫组化EnV ision两步法检测60例鼻咽癌标本及15例正常鼻咽黏膜中Ptch1/2蛋白的表达,比较Ptch1与Ptch2表达的差异,分析Ptch1和(或)Ptch2与鼻咽癌患者临床病理特征之间的关系。结果鼻咽癌组织中Ptch1/2蛋白表达的综合评分分别为4.70±3.59、3.58±3.47,明显高于正常鼻咽黏膜上皮组织的2.93±1.98、2.07±1.10(均P<0.05);肿瘤或正常鼻咽黏膜上皮组织中Ptch1的表达水平均稍高于Ptch2;不同Ptch1及(或)Ptch2蛋白表达水平的鼻咽癌临床病理特征的比较均差异无统计学意义(P>0.05);Ptch1与Ptch2蛋白表达在正常组织中呈显著相关(r=0.592,P<0.05),而在肿瘤组织中则无相关性(r=0.114,P>0.05)。结论鼻咽癌组织中Ptch1/2表达上调可能共同参与鼻咽癌的发生及发展,其中尤以Ptch1占主导地位。
Objective To investigate the correlation of the expression of the Hedgehog signaling pathway mem- bers Ptch1 and Ptch2 (Ptch1/Ptch2) in nasopharyngeal carcinoma ( NPC ). Methods The expression of Ptchl and Ptch2 in 60 NPC biopsy specimens and 15 normal human nasopharyngeal epithelia tissues was assessed by immunohisto- chemistry. The correlation between the levels of Ptchl and Ptch2, and the correlation between the clinicopathological fea- tures and the expression of Ptch1/Ptch2 were analyzed. Results The expression of Ptchl and Ptch2 was observed in NPC specimens, significantly higher than that in normal human nasopharyngeal epithelial samples ( Ptchl : 4. 70 ± 3.59 vs. 2. 93 ±1.98, Ptch2 : 3.58 ± 3.47 vs. 2. 07±1.10, P 〈 0. 05). But the expression level of Ptchl or Ptch2 was not signif- icantly correlated with clinieopathological characteristics ( P 〉 0. 05 ). The level of Ptchl expression was slightly higher than Ptch2 in carcinoma and normal nasopharyngeal epithelia ( P 〉 0. 05 ). There was significant correlation between Ptchl and Ptch2 expression in normal nasopharyngeal epithelia ( r = 0. 592, P 〈 0. 05 ), though not in NPC tissues ( r = 0. 114, P 〉 0. 05 ). Conclusion It is suggested that the up - regulation of Ptch1/Ptch2 expression may be involved in the occur- rence and progression of NPC, especially Ptchl.
