正交优化微波辅助提取马铃薯皮渣多酚工艺及抑菌研究 被引量:5

Study on Optimization of Microwave-assisted Extraction of Polyphenols from Potato Peels by Orthogonal Design and Their Antimicrobial Activity

摘要 为了探讨微波法提取马铃薯皮渣中多酚的最佳工艺条件及其体外抑菌效果,试验通过正交设计和二次多项式逐步回归分析,以优化出最佳提取工艺,并利用纸片琼脂扩散法对其抑菌效果进行测定。结果表明,微波辅助提取马铃薯皮渣多酚的最佳工艺条件为:浸提剂乙醇浓度为55%、料液比为1∶25(g/m L)、微波功率50 W、微波时间40 s,多酚提取量为3.02 mg/g;抑菌效果分析显示,从马铃薯皮渣中提取得到的多酚均能够抑制金黄色葡萄球菌、大肠杆菌和枯草芽孢杆菌,抑菌的最低质量浓度均为10 mg/m L,在这3种菌中,大肠杆菌对其最为敏感。 To research the method of extracting polyphenols from potato peels and their antimicrobial activity, the optimum extraction technology was determined through orthogonal design and two degree polynomial regression analysis, as well as, the bacteriostasis tests were conducted by paper-disc agar-diffusion method. The results showed the ethanol concentration was 55%, the ratio of material to liquid was 1 ∶25(g/m L), microwave power was 50 W, microwave time was 40 s, under this condition,polyphenols extraction rate was up to 3.02 mg/g. Bacteriostasis experiments showed that the polyphenols from potato peels had antibacterial activity against Staphylococcus aureus, Escherichia coli and Bacillus subtulis and the minimal inhibitory concentration was 10 mg/m L. Among the 3 bacteria, it had the most significant inhibition effect for Escherichia coli.
