摘要
目的建立UPLC-MS/MS法结合模式识别同时测定芪参益气滴丸(黄芪、丹参、三七等)中丹参素、原儿茶酸、原儿茶醛、咖啡酸、迷迭香酸、丹酚酸B、丹酚酸A、丹参酮Ⅰ、隐丹参酮、丹参酮ⅡA、熊果酸的含有量。方法该药物50%甲醇提取液的分析采用ACQUITY UPLCBEH C18色谱柱(2.1 mm×50 mm,1.8μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量0.2 m L/min;柱温40℃。再采用SIMCA14.0软件进行模式识别。结果 11种成分在各自范围内均呈良好的线性关系(r≥0.999 6),平均加样回收率98.45%~101.38%,RSD 1.70%~2.40%。10批样品质量偏差均在2 std(标准偏差)范围内。结论该方法简便、灵敏、准确,可用于芪参益气滴丸的质量控制。
AIM To establish a UPLC-MS/MS combined with pattern recognition method for the simultaneous content determination of danshensu,protocatechuic acid,protocatechuic aldehyde,caffeic acid,rosmarinic,salvianolic acid B,salvianolic acid A,tanshinone I,cryptotanshinone,tanshinone IIA and ursolic acid in Qishen Yiqi Dropping Pills (Astragali Radix,Salviae miltiorrhizae Radix et Rhizoma,Notoginseng Radix et Rhizoma,etc.).METHODS The analysis of 50% methanol extract of this drug was performed on a 40 ℃ thermostatic ACQUITY UPLCBEH C18column (2.1 mm × 50 mm,1.8 μm),with the mobile phase comprising of acetonitrile-0.1%formic acid flowing at 0.2 m L/min in a gradient elution manner.Then SIMCA14.0 software was adopted in the pattern recognition.RESULTS Eleven constituents showed good linear relationships within their own ranges (r≥0.999 6),whose average recoveries were 98.45%-101.38% with the RSDs of 1.70%-2.40%.The quality deviations of ten batches of samples were all within 2 std (standard deviation).CONCLUSION This simple,sensitive and accurate method can be used for the quality control of Qishen Yiqi Dropping Pills.
