七叶皂苷冻干粉针对缺血性脑水肿小鼠的治疗作用及机制研究 被引量:4

摘要 目的:观察七叶皂苷钠冻干粉对缺血性脑水肿小鼠的治疗作用及机制研究。方法:制备小鼠脑缺血模型,将实验小鼠随机分为4组,空白对照组、模型组、地塞米松组、七叶皂苷钠组。七叶皂苷钠组于尾静脉注射七叶皂苷钠3.6 mg/kg,地塞米松组按等浓度的地塞米松磷酸钠注射液尾静脉注射,模型组及空白对照组给予等容积的0.9%生理盐水,每天1次,连续给药7 d。干湿称重法测定各组大鼠脑组织含水量,ELISA法测定脑组织IL-1β和ET含量。结果:与空白组比较,模型组小鼠脑组织含水量显著升高,脑组织IL-1β和ET含量均升高,具有统计学意义(P<0.01);与模型组比较,地塞米松组、七叶皂苷钠组小鼠脑组织含水量降低,脑组织IL-1β和ET含量均降低,具有统计学意义(P<0.01)。结论:七叶皂苷钠冻干粉能减少缺血性脑水肿小鼠出血后脑水肿程度,改善脑组织病理变化,其作用机制与降低脑组织IL-1β和ET含量有关。 Objective : To study the therapeutic effect and mechanism of Aescine producing injection in mice with ischemic brain edema. Methods : Prepare mice cerebral ischemia model. The experimental mice were randomly divided into four groups, control group, model group, dexamethasone group and Aescine group. Aescine groups were injected through tail vein 3.6 mg/kg, once a day, for seven days. Dexamethasone group were given equal dose dexamethasone. Model and control groups were given equal dose 0.9% physiological saline, once a day, for seven days. Determine water content in brain tissue of rats by wet and dry weight method. Determine IL - 1 and ET contents in brain tissue by ELISA. Results : Compared with the control group, model group's water content of brain tissue increased. The contents of IL - 1β and ET were both increased ( P 〈 0. 01 ). Compared with the model group, Dexamethasone and Aescine groups' water content of brain tissue decreased and the contents of IL - 1β and ET were both decreased(P 〈 0. 01 ). Conclusion : Aescine producing injection can reduce the degree of brain edema in mice with ischemic brain edema and improve the pathological changes of brain tissue. The mechanism of action is related to the decrease of the contents of IL - 1 β and ET in brain tissue.
