静脉留置导管相关纤维蛋白鞘模型的建立 被引量:6

摘要 目的建立静脉留置导管相关纤维蛋白鞘动物模型。方法选用46只家兔,预实验6只随机均分为2组(留置6h、12h组);正式实验40只随机均分为5组(留置12h、1d、2d、3d、4d组)。采用留置针置管于兔耳缘静脉,记录每次滴注造模药物所需时间,留取不同时相的纤维蛋白鞘标本,行HE、Masson染色以观察其组织形态学变化。结果预实验:留置6h时,并无纤维蛋白鞘形成,而留置12h时,开始形成纤维蛋白鞘;正式实验:不同组组间第一、二、三、四次滴注造模药物所需时间比较,差异均无统计学意义(P>0.05),而不同组组内第一、二、三、四次滴注造模药物所需时间比较,差异均具有统计学意义(P<0.001);不同组纤维蛋白鞘面积占静脉管腔面积百分比比较,差异具有统计学意义(P<0.001);分析HE、Masson染色结果,确定纤维蛋白鞘包含有血栓、红细胞、纤维素、胶原纤维和某些细胞成分。结论此方法建立的模型可靠稳定、成功率较高,符合导管相关纤维蛋白鞘的生物学特性。 ter. Methods Objective Forty-six Aim to establish an animal model of the fibrin sheath in the vein indwelling cathe rabbits were chosen, six rabbits involved in pre-experiment were randomly and e qually divided into two groups (indwelling 6 h and 12 h groups), forty rabbits involved in formal experiment were randomly and equally divided into five groups (indwelling 12 h, 1 d, 2 d, 3 d, 4 d groups). Models were made by using indwelling needle for placement tube on rabbit ear edge vein, recorded each dripping drug time needed for building models, collected fibrin sheath samples at different phase at the same time, the HE staining and Masson staining were performed to observe the histomorphological changes. Results Pre-experiment results: indwelling 6 h, no formation of fibrin sheath was found, indwelling 12 h, fibrin sheath began to develop. Formal experiment results: horizontal comparing the dripping drug time needed for completing models at the first, the second, the third, and the fourth times among different groups showed that there was no statistically significant difference ( P 〉0.05), and the longitudinal intra group comparing the dripping drug time needed for completing models at the first, the second, the third, and the fourth times among different groups showed that there was statistically significant difference ( P 〈0. 001). Comparison the ratio of the fibrin sheath area to the venous luminal area among different groups showed statistically significant difference ( P 〈0. 001). Analyzing the results of the HE and Masson staining confirmed that the fibrin sheath contained blood clots, red blood cells, cellulose, collagen fibers and certain cell components. Conclusion The venous indwelling catheter re lated fibrin sheath model developed by the mentioned above rate of modeling, and the model is in accordance with the method is reliable and stable, with high successful biological characteristics of catheter related fibrin
