摘要
建立可快速准确测定海洋小单孢菌发酵液中Rakicidin B含量的HPLC方法。采用液相色谱系统(HPLC)Agilent 1260 infinifty,色谱柱:Syncronis C18(5μ,4.6×250mm)。流动相:乙睛:水=85:15,检测波长为260 nm,柱温为30℃。结果:Rakicidin B浓度范围在30~600ug/ml线性良好,相关系数为0.9998(n=7);发酵液样品的加样回收率为99.2~105%,平均回收率101.57%,回收率相对标准偏差RSD为2.2049%(<3%),方法的精密度RSD为0.444%。结论:方法准确可靠,可以用于Rakicidin B发酵、提炼及纯化过程的含量测定。
Object:Develop a HPLC method for the determination of Rakicidin B in fermentation broth of marine Micromonospora sp. Methods: Using HPLC Agilent 1260 infinifty,Syncronis C18(5μ,4.6×250mm),the mobile phase was acetonitrile : water =85:15 at a flow rate of 1.0m L.min-1and detection wavelength was 260 nm,column temperature was30 ℃. Results:The calibration curve was linear over the range of 30-600ug/ml with the correlation 0.9998(n=7). The average recovery of daptomycin was 101.57%(RSD=0.444%). Conclusion:The method is accurate,simple with good reproducibility,which is suitable for the determination of Rakicidin B in fermentation broth.
