摘要
目的:探索灯台七总皂苷提取工艺优化及其抗肿瘤活性研究。方法:采用响应面优化法优化灯台七中总皂苷的提取工艺;使用最佳的提取工艺提取、比较了陕西地区10批不同产地灯台七茎叶与根茎总皂苷含量差异;使用MTT法分别对不同产地灯台七总皂苷进行了抗肿瘤活性测试。结果:最佳工艺条件为提取时间60min、提取次数3次、乙醇浓度65%、液料比6∶1。总皂苷含量:茎叶>根茎;抗肿瘤活性测试发现不同来源灯台七总皂苷均有非常好的抑制SW620、HT-29生长的作用;同时MTT实验表明,70%醇部位的皂苷对SW620、HT-29细胞的生长抑制作用最强。结论:该工艺条件可为灯台七总皂苷的工业生产提供依据。
Objective:To explore extraction of total saponins from Paris Polyphylla and its antitumor activity.Methods:To optimize the extracting process of the total saponins of Paris Polyphylla.The best extraction process was used to extract and compare the contents of total saponins in stems and leaves and rhizome of 10 batches of different origin Paris Polyphylla in Shaanxi province.MTT assay was used to test the antitumor activity of the total saponins of the different origin Paris Polyphylla.Results:The optimum conditions of extraction were as follows.Eextraction time was 60 min extraction times was 3,ethanol concentration was 65%,solvent volume times was 6∶1.Compared to the rhizome,total saponin content of stem and leaf are more high.Antitumor activity test found that total saponins of different sources ParisPolyphylla all have a very good inhibition for SW620 and HT-29 growth.MTT assay showed that 70%alcohol saponin had the strongest inhibitory effect on SW620 and HT-29 cells.Conclusion:The process conditions can provide the basis for the industrial production of the ParisPolyphylla total saponin.
