KCNJ11基因多态与糖尿病前期人群运动干预敏感性的相关性研究 被引量:3

Correlation analysis between KCNJ11 gene polymorphism and susceptibilities to exercise intervention in prediabetes

摘要 目的研究钾离子通道蛋白KCNJ11基因区的3个标签单核苷酸多态性位点(SNP),及其组成的单体型与糖尿病前期人群运动干预前后糖脂代谢相关指标变化的关系,探讨其作为糖代谢异常人群对运动干预敏感的分子标记。方法对70名糖尿病前期人群进行为期12周的有氧耐力运动干预,测定其干预前后糖脂代谢相关生化指标。采用MALDI-TOF MS方法解析受试者基因组DNA多态位点基因型,分析干预前后相关指标的变化与单个位点及多个位点组成的单体型的相关性。结果 rs5219位点的CC基因型与HOMA-IR、TC、TG指标的下降、与HDL-C指标的升高相关,rs2285676位点的GG基因型与FPG、HbA1c、HOMA-IR、TC的下降相关;rs5219-rs2285676-rs5218位点组成的T-A-G单体型与HOMA-IR的下降相关,C-G-A单体型与TC指标的下降相关,rs5219-rs5218位点组成的C-A单体型与FPG的下降相关,rs2285676-rs5218位点组成的G-G单体型与HDL-C的升高相关。结论KCNJ11基因rs5219位点的CC基因型、rs2285676位点的GG基因型及部分单体型与运动干预前后糖脂代谢的不同指标变化有相关性,可作为评价糖尿病前期人群对运动干预敏感性的参考指标。 Objective To investigate the relationship between three SNPs and their haplotypes in KCNJ11 gene region and the changes of glucose and lipid metabolism indicators after aerobic endurance exercise intervention. The aim is to probe into the feasibility of using them as sensitive molecular markers of exercise intervention susceptibilities in prediabetic population. Methods A total of 70 subjects with prediabetes were intervented by aerobic endurance exercise for 12 weeks. The biochemical indexes related to glucose and lipid metabolism were measured before and after intervention. The genotypes of SNPs were analyzed by MALDI-TOF MS method in all subjects. The correlations between changes of indexes and each SNP,as well as haplotypes were analyzed. Results CC genotype of rs5219 was associated with the decrease of HOMA-IR, TC and TG, and with the increase of HDL-C. GG genotype of rs2285676 was related with the decline of FPG, HbA1 c, HOMA-IR and TC. TAG and CCA haplotype (rs5219-rs2285676- rs5218) were associated with the decrease of HOMA-IR and TC respectively. CA haplotype (rs5219- rs5218) and GG haplotype (rs2285676 rs5218) were associated with the decrease of FPG and the increase of HDL-C respectively. Conclusion The CC genotype (rs5219), GG genot.ype (rs2285676), and some haplotypes of KCNJll gene are associated with the changes of glucose and lipid metabolism indexes after exercise intervention. They may serve as sensitivity molecular markers for evaluation of responses to exercise intervention in subjects with prediabetes.
