摘要
目的:探讨白介素17F(IL-17F)基因多态性rs763780和rs2397084与类风湿关节炎(RA)易感性的关系。方法:计算机检索中国学术期刊全文数据库、万方数据库、中国生物医学文献数据库、Pubmed、Embase以及Web of Science中已发表的与RA和IL-17F基因多态性相关文献,进行Meta分析。结果:纳入的5项研究共收集RA患者1 174例(其中rs763780含1 174例,rs2397084含985例),正常对照966例(rs763780含966例,rs2397084含766例)。Meta分析结果显示,rs763780与RA密切相关(C vs.T:OR=1.74,95%CI=1.01-2.98,P=0.04;CC vs.TT:OR=3.17,95%CI=1.21-8.35,P=0.02;CT vs.TT:OR=1.80,95%CI=1.21-2.66,P=0.004;CC+CT vs.TT:OR=1.96,95%CI=1.35-2.83,P<0.001;CC vs.CT+TT:OR=2.97,95%CI=1.13-2.66,P=0.03)。根据人种进行亚组分析发现,在欧洲人群中rs763780与RA密切相关,差异具有统计学意义。未发现rs2397084与RA之间的相关性。结论:IL-17F rs763780是RA的危险因素,IL-17F rs2397084与RA易感性是否有关仍不明确。
Objective :To explore the association between rheumatoid arthritis(RA)and two polymorphisms of interleukin-17 (IL-17F)rs 763780 and rs 2397084. Methods: A systematic search of relevant studies was conducted in China National Knowledge In-frastructure, Wanfang Chinese Periodical Database,China Biology Medical Literature Database, Pubmed, Embase and Web of Science. Results: A total of 5 case-control studies including 1 174 RA cases and 966 health controls for rs 763780 and 985 RA cases and 766 health controls for rs 2397084 were included in this Meta- analysis. The results showed that rs 763780 was significantly associated with increased susceptibility to RA(C vs. T: OR=1. 74,95% CI =1. 01-2. 98, P = 0. 04;CC vs. TT:OR = 3. 17,95% CI =1. 21-8. 35, P = 0.02;CT vs.TT:OR = 1.80,95% CI =1.21-2.66, P = 0.004;CC + CT vs.TT:OR = 1.96,95% Cl = 1.35-2.83, P〈0.001;CC vs. CT+ TT:OR = 2. 97,95% CI =1. 13-2. 66, P = 0.03). In subgroup analysis based on ethnicity, rs 763780 was positively correlated with RA in Europeans. No significant association was found between rs 2397084 and RA. Conclusion: The present study revealed that IL-17F rs 763780 polymorphism is a risk factor of RA,while the association of rs 2397084 with RA remains uncertain.
