红叶腺柳继代培养研究

摘要 以红叶腺柳(Salix chaenomeloides‘Variegata’)带芽茎段诱导出的侧芽为试验材料,研究了不同细胞分裂素、生长素、添加物、抗褐化剂及降低激素浓度等因素对红叶腺柳试管苗继代增殖及高生长的影响。结果表明,适宜的丛生芽增殖培养基为:WPM+6-BA1.0mg·L^(-1)+NAA0.1mg·L^(-1);利于试管苗伸长培养基为:WPM+6-BA0.5mg·L^(-1)+NAA0.1mg·L^(-1)+活性炭1.0g·L^(-1),添加抗褐化剂活性炭,并采用1.0mg·L^(-1)6-BA与0.5mg·L^(-1)6-BA交替使用的方法 ,在保持红叶腺柳高繁殖率的同时促进试管苗高生长,抑制茎尖坏死、玻璃化、褐化等不良现象。 Abstract Lateral buds of Salix chaenomeloides 'Variegata' were used as materials to study effect of different kinds and concentration of cytokinins,auxins,additives,browning inhibitors and reduced plant growth regulators concentration on shoot multiplication and elongation. The optimum medium for shoot multiplication is: WPM +6 -BA1.0mg·L^-1 +NAA0. 1mg·L^-1 ; The optimum medium for shoot Elongation is:WPM +6 -BA0.5mg·L^-1+NAA0.1mg ·L^-1+active carbonl.0g ·L^-1.Add active carbon, combined with using 1.0mg·L^-16-BA and 0.5mg ·L^-16-BA alternately, which can Maintain a high multiplication value, shoot elongation without apical necrosis,vitrification and browning.
