摘要
通过分离、纯化和鉴定江西省苦瓜枯萎病致病菌株,探讨适用于江西地区苦瓜苗期人工接种的抗性鉴定方法,对36个苦瓜品种进行抗性鉴定,筛选高抗资源,以期为抗性育种提供科学的理论基础。采用室内筛选和田间试验相结合的方法。从江西省苦瓜枯萎病病株茎基部分离、纯化了3株致病力较强的尖孢镰刀菌(Fusarium oxysporum),通过形态观测、致病力检测和rRNA-ITS序列分析,初步确定该病是由尖孢镰刀菌苦瓜专化型(F.oxysporum f.sp.momdicae)。优化了适用于江西地区苦瓜苗期人工接种方法,在此基础上,采用浸根移栽法对36个苦瓜品种进行枯萎病抗性(Fusarium oxysporum f.sp.)鉴定,发现具有高抗枯萎病特性材料6份,抗病材料11份,中抗材料4份,高感材料9份。
In order to provide a scientific basis for the anti-wilt breeding program of fusarium wilt of in bitter gourd in Jiangxi Province, it is very important to screen and identify the pathogen which causes fusarium wilt.The resistance identification technology was investigated with inbred lines presenting different degrees of field resistance.A lab-screening and field experiment were carried out to identify the pathogen.Three pathogen-ic strains were obtained from the diseased plants. The pathogen was isolated and characterized based on the morphological features,pathogenicity test, and sequence analysis of internal transcribed spacer region of the ribosomal DNA(rRNA- ITS) .The results showed that the fusarium wilt was caused by fusariumoxysporum f. sp. momdicae. Then a total of 36 Momordicacharantia varieties ( or inbred lines) were evaluated by root-soaking transplant inocultaton lor resistance against Fusarium wilt,6 materials of high resistance( HR) ,11 ol resistance (R) ,4 of moderate resistance(MR)and 9 of high supceptibility(HS)were found.
