摘要
目的:观察百诃清金颗粒对气道高反应性大鼠气道炎症的干预作用。方法:将48只SD大鼠随机分为空白组、模型组、地塞米松组、百诃清金颗粒低、中、高剂量组,每组8只;采用卵白蛋白致敏,复制气道高反应性大鼠模型。按实验计划干预后,记录各组支气管肺泡灌洗液(BALF)中白细胞总数及分类计数;检测白介素-4(IL-4)、白介素-6(IL-6)、干扰素-γ(IFN-γ)、肿瘤坏死因子(TNF-α)的含量;观察肺组织HE染色病理学特点。结果:百诃清金颗粒高、中剂量组可明显降低气道高反应大鼠BALF中白细胞、嗜酸性粒细胞(P<0.001)及中性粒细胞数量(P<0.05),并减轻肺组织炎症损伤。与空白组相比,模型组IL-4、IL-6、TNF-α水平显著升高(P<0.001),IFN-γ水平显著下降(P<0.001)。高剂量组可显著降低IL-4(P<0.001)、IL-6、TNF-α(P<0.01)表达,升高IFN-γ(P<0.01)水平,效果与地米组相当;中、低剂量组IL-4、TNF-α水平较虽明显低于模型组,但差异无统计学意义。结论:百诃清金颗粒可抑制气道高反应性大鼠的气道炎症反应,降低气道高反应性,且呈明显量效关系;这可能是其治疗感染后咳嗽等以气道反应性增高为病理表现疾病的部分作用机制。
Objective: To observe the effect of Baihe Qingjin Granule on lung inflammation in rats with airway hyperrespons- iveness. Methods: Forty-eight SD rats were randomly into 6 groups including the normal group, model group, dexamethasone group and three Baihe Qingjin Granule groups at dose of 21, 10. 5, 5. 25' g/kg with 8 rats in each groups. Copying the rats model of airway hyperresponsiveness which induced by Ovalbumin. According to the plan to gavage, recorded the total and differ- ential leukocyte count in bronchoalveolar lavage fluid (BALF), detection of interleukin-4 (IL-4), interleukin-6 (IL-6) and interferon gamma (IFN-γ), tumor necrosis factor alpha (TNF-α) in BALF; observing the lung tissue pathological features un- der the HE staining. Results: Compared to the model group, Baihe Qingjin Granule in high and middle dose group can signifi- cantly reduce the white blood cells, eosinophils (P 〈 0. 001 ) and the number of neutrophils (P 〈 0. 05) in rats of BALF with AHR, and alleviated the inflammatory injury in lung tissue. Compared with the normal group, the levels of IL-4, IL-6 and TNF -α in the model group were significantly increased (P 〈 0. 001 ), and the levels of IFN-γ was obviously decreased (P 〈 0. 001 ) ; The high dose group could significantly reduce the expression of IL-4 ( P 〈 0. 001 ), IL-6, TNF-α ( P 〈 0. 01 ), and increased the level of IFN-γ(P 〈 0. 01), which was similar to dexamethasone group. The levels of IL-4 and TNF-α in the middle and the low dose group were lower than those in the model group, but the difference was no statistically significant. Con- clusion: Baihe Qingjin Granule can significantly inhibit airway inflammatory in rats with airway hyperresponsiveness and showes marked dose-effect relationship, which is the part of the mechanism of treatment of cough after infection.
