核蛋白因子泛素样含锌指状结构域和环指域1在乳腺癌组织的表达及意义

摘要 目的观察核蛋白因子泛素样含锌指状结构域和环指域1(UHRF1)在乳腺癌及正常乳腺组织中的表达。 方法采用实时定量反转录聚合酶链反应(RT-qPCR)法及Western blot法检测40例乳腺癌组织及30例正常乳腺组织中UHRF1基因的mRNA的表达及其在蛋白水平的表达,并分析UHRF1的表达与临床相关病理参数间的关系。结果UHRF1 mRNA在乳腺癌组织中的表达量(1.34±0.03)明显高于正常乳腺组织(0.61±0.03);其蛋白表达明显高于正常乳腺组织。乳腺癌组织中UHRF1的表达水平与临床分期及是否有淋巴结转移明显相关。结论UHRF1的阳性表达与乳腺癌的发生有关。 Objective To observe the expression and significance of ubiquitin - like with PHD and ring finger domain 1 ( UHRF1 ) in breast cancer and adjacent normal tissue. Methods The expression level of UHRF1 mRNA and protein was analyzed by real - time quantitative reverse transcriptase - polymerase chain reaction (RT- qPCR) and Western blotting, respectively. Relationships between UHRF1 protein positive expression and clinicopathological characteristics were analyzed. Results The UHRF1 mRNA expression level in breast cancer ( 1.34 -+ 0. 03 ) was higher than in adjacent normal tissue (0. 61 _+ 0. 03 ). And the UHRF1 protein expression level in breast cancer was also higher than in adjacent normal tissue. The UHRF1 protein positive expression in breast cancer tissue was significantly correlated with clinical sta- ges and lymphatic metastasis. Conclusion UHRF1 protein positive expression is correlated to the occurrence of breast cancer.
