归芪多糖对衰老大鼠肾组织氧化损伤作用的研究 被引量:7

摘要 目的:研究归芪多糖对D-半乳糖所致衰老大鼠肾组织氧化损伤的影响。方法:通过水提醇沉法从一定比例的当归和黄芪中提取出归芪多糖,并用苯酚-硫酸法进行多糖含量测定;除正常对照组外其余各组均采用D-半乳糖(0.15 g/kg/d)皮下注射建立大鼠衰老模型;以不同剂量的归芪多糖(2.0g/kg和0.5 g/kg)给大鼠连续灌胃42 d后,测定其肾组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活力、谷胱甘肽(GSH)含量、丙二醛(MDA)含量和蛋白质羰基含量。结果:归芪多糖可增强衰老大鼠肾组织中的SOD、GSH-PX活力和GSH含量,显著降低MDA和蛋白羰基含量。结论:归芪多糖可保护D-半乳糖所致衰老大鼠肾组织的氧化损伤,从而起到保护肾脏衰老的作用。 Objective:To study the effect of Guiqi polysaccharide on oxidative damage of kidney tissue in D-galactose-induced aging rats.Methods:We extracted polysaccharide from a percentage of angelica and astragalus membranaceus through the water extraction and alcohol precipitation method,and determined the content of polysaccharide with phenol sulfuric acid method.D-galactose was subcutaneous injected at a dose of 0.15 g/kg body weight every day except the normal control group.Rats of the low-and high-dose Guiqi polysaccharide( 2.0 g/kg and 0.5 g/kg) were orally administrated daily for 42 days,respectively.Then superoxide dismutase( SOD) activity,the content of glutathione peroxidase( GSH-PX),glutathione( GSH),malondialdehyde( MDA) content,protein carbonyl of kidney were determined,respectively.Results:Guiqi polysaccharides could enhance SOD,GSH-PX,GSH enzyme activity in renal tissue of aging rats,and it could significantly decrease the content of MDA and protein carbonyl.Conclusion:Guiqi polysaccharide can reduce free radical damage to the kidney of aging rats and protect the normal kidney from senescence.
