甲基苯丙胺对大鼠海马TNF-α、IL-1β及CD22表达的影响 被引量:2

摘要 目的研究甲基苯丙胺(methamphetamine,MA)对大鼠海马的肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(Interleukin-1β,IL-1β及CD22的影响.方法通过连续腹腔注射MA15 mg/(kg.次)1 d、3 d、5 d,每天2次,建立正常、1 d、3 d、5 d组MA诱导中毒大鼠模型,每次给药后,各组结合刻板行为观察并评分;应用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测实验大鼠海马TNF-α与IL-1β的表达含量;Western blot检测大鼠海马CD22表达含量.结果与正常组大鼠比较,1 d、3 d、5 d组大鼠海马TNF-α与IL-1β的表达含量呈递增趋势,且5 d组显著升高(P<0.01),CD22表达含量亦呈递增趋势,且5 d组显著升高(P<0.01).结论 MA可使大鼠海马TNF-α与IL-1β以及CD22蛋白表达明显增加. Objective To study the effect of methamphetamine on the expression of TNF-α, IL- β and CD22 in rats hippocampus. Method MA (15 mg/kg ) was daily administrated (I.P) twice a day for lday, 3days and 5days and normal control group, one-day group, three-day group and five-day group were established. After injection of MA, the stereotyped behavior activities in rats were scored. ELISA method was applied to detect the changes of TNF alpha and IL -1 beta in experimental rats. Western blot was used to detect the expression of CD22. Results Compared with normal control group, the expression of TNF alpha and IL -lbeta increased in different MA treated groups, and there was a significant difference in 5-day group (P 〈 0.01) ; the expression of CD22 increased spirally and there was a significant difference in 5 days group (P 〈 0.01) Conclusion MA administration resulted in significant increase in TNF alpha, IL -lbeta and CD22 expression in rats hippocampus,
