摘要
目的探讨瘤果黑种草子总黄酮(TFNG)对RAW264.7细胞及豚鼠气管平滑肌的作用。方法采用酶联免疫吸附法(ELISA)、Griess Reagent法检测高、中、低剂量(20,40,80μg/mL)TFNG对脂多糖(LPS)体外诱导的RAW264.7细胞分泌白细胞介素8(IL-8)、IL-6、一氧化氮(NO)的影响;采用离体气管恒温灌流的方法,制备豚鼠气管螺旋条标本,通过观察其张力变化考察1,2,4 g/L不同剂量TFNG和10 g/L水煎液对张乙酰胆碱(ACh)和组胺(His)所致痉挛离体豚鼠气管平滑肌解痉作用。结果 TFNG低于100μg/mL时对RAW264.7细胞无明显的毒性作用;与LPS组比较,TFNG低、中、高剂量(20,40,80μg/mL)均能抑制RAW264.7细胞过量分泌IL-6,IL-8,NO(P<0.05),且呈一定量效关系;TFNG低、中、高剂量组(1,2,4 g/L)可明显舒张ACh及His引起的豚鼠支气管平滑肌痉挛,差异有统计学意义(P<0.01)。结论 TFNG具有一定的体外抗炎及支气管解痉功能。
Objective To study the effect of total flavones from Nigella Glandulifera( TFNG) on RAW264. 7 cells and guinea pig tracheal smooth muscle. Methods The high, medium and low dose( 20, 40, 80 μg/mL) of TFNG was used to detect the secretion of interleukin-8 in RAW264. 7 cells induced by lipopolysaccharide( LPS) by enzyme-linked immunosorbent assay( ELISA) and Griess Reagent method. IL-8, IL-6 and nitric oxide( NO) were measured. The tracheal spiral specimens of guinea pigs were prepared by the method of constant-temperature perfusion with tracheal trachea. The changes of the tension were observed by 1,2,4 g/L dose of TFNG and 10 g/L decoction on antipruritic effects of guinea pig tracheal smooth muscle induced by tacrolipotentin( ACh) and histamine( His).Results TFNG had no significant toxic effect on RAW264. 7 cells at less than 100 μg/mL. Compared with LPS group,the TFNG low,medium and high dose group(20,40,80 μg/mL) could inhibit RAW264. 7 cells and the levels of IL-6,IL-8 and NO( P 〈0. 05),and there was a significant dose-dependent relationship. The TENG low, medium and high dose group( 1,2,4 g/L) could significantly relax the bronchial smooth muscle spasms of guinea pig induced by ACh and His,the difference was statistically significant( P〈 0. 01).Conclusion TFNG has some anti-inflammatory and bronchial antispasmodic effects in vitro.
