茶皂素对奶牛免疫功能的影响 被引量:14

Effects of Tea Saponin on Immune Function of Dairy Cows

摘要 本试验旨在研究茶皂素对荷斯坦奶牛的血清免疫相关细胞因子含量及其mRNA表达量的影响。试验采用4×4拉丁方设计,选取4头瘤胃瘘管奶牛,每头奶牛分别灌注0(对照)、15、30、45 g/d的茶皂素,预试期14 d,正试期21 d,共4期。采用酶联免疫吸附剂测定(ELISA)法试剂盒测定血清中免疫球蛋白(Ig)、白细胞介素(IL)和肿瘤坏死因子-α(TNF-α)的含量,采用实时荧光定量PCR方法测定淋巴细胞IL-1、IL-6、IL-10和TNF-αmRNA表达量。结果显示:1)30 g/d茶皂素组奶牛血清Ig M含量显著高于对照组(P<0.05)。2)15 g/d茶皂素组奶牛血清IL-1含量显著高于对照组和45 g/d茶皂素组(P<0.05)。3)与对照组相比,15、30、45 g/d茶皂素组奶牛血清IL-1、IL-6、IL-10、TNF-αmRNA表达量均无显著差异(P>0.05),但45 g/d茶皂素组可提高以上细胞因子mRNA表达量。由此可见,茶皂素可提高奶牛血清中Ig及免疫相关细胞因子含量,并提高细胞因子IL-6 mRNA表达量,表明茶皂素可提高奶牛免疫功能。 This experiment was conducted to study the effects of tea saponin on serum immune related cyto- kines contents and their mRNA expression levels of Holstein cows. Four rumen fistulated cows were used fol- lowing a 4x4 Latin square design with different infusion concentrations of tea saponin U 0 (control), 15, 30, 45 g/dl. The experiment had 4 periods, and each period consisted of 14 d per feeding period and 7 d sampling period. Enzyme-linked immunosorbent assay (ELISA) method kit was used to measure the contents of immu- noglobulins, interleukin and tumor necrosis factor alpha (TNF-a) in serum. Quantitative real-time PCR was used to measure the mRNA expression levels of interleukin-1 (IL-l), interleukin-6 (IL-6), interleukin-10 (IL-10) and TNF-a. The results showed as follows: 1 ) the serum immunoglobulin M (IgM) content of dairy cow in 30 g/d tea saponin group was significantly higher than that in control group (P〈0.05). 2 ) The serum IL-1 content in 15 g/d tea saponin group was significantly higher than that in control group and 45 g/d tea sap- onin group (P〈0.05). 3) Compared with the control group, the mRNA expression levels of IL-1, IL-10, IL- 6 and TNF-oL in 15, 30 and 45 g/d tea saponin groups were not significantly different (P〉0.05), but the mR- NA expression levels of above cytokines in 45 g/d tea saponin group was increased. In conclusion, tea saponin can improve the contents of immunoglobulins and immune related cytokines in serum, as well as the mRNA ex- pression level of cytokine IL-6, which indicate that tea saponin can improve the immune function of dairy cows.
