木薯抗草甘膦突变株系离体筛选

Screening for Resistant Glyphosate Somaclonal Variation of Cassava to Glyphosate in Vitro

摘要 以木薯(Manihot esculenta Crantz)品种桂热-6、桂热-4试管苗叶片为材料,研究草甘膦对愈伤组织增殖与分化的影响,以建立抗草甘膦的木薯离体筛选体系。结果表明,桂热-6、桂热-4幼叶愈伤组织诱导与增殖的适宜配方分别为改良B5+2,4-D 0.50~0.75 mg/L、改良B5+2,4-D 0.75 mg/L;愈伤组织分化不定芽的适宜配方分别为改良B5+TDZ 0.75 mg/L、改良B5+TDZ 0.75~1.00 mg/L。愈伤组织增殖过程中,桂热-6、桂热-4愈伤死亡率达到50%左右所需的草甘膦浓度分别为800、600 mg/L;愈伤组织分化过程中,桂热-6、桂热-4愈伤组织死亡率达到50%左右所需的草甘膦浓度分别为600、400 mg/L。在含草甘膦的愈伤组织增殖培养基、愈伤组织分化培养上分二步筛选获得了桂热-6木薯抗草甘膦无性系。温室幼苗喷施草甘膦试验表明,抗性无性系幼苗对草甘膦的抗性得到增强,研究结果可为木薯无性系变异育种研究提供参考。 For screening resistant somaclonal variation of cassava to Glyphosate in vitro, the study was made to clarify the effects of glyphosate on proliferation and differentiation of callus from plantlet young leaves in cassava(Manihot esculenta Crantz)species Gui Re-6 and Gui Re-4. The optimum induction and proliferation medium of callus were modified B5 medium supplemented with 0.50~0.75 mg/L 2,4-D for leaves strips from Gui Re-6 and 0.75 mg/L 2,4-D for leaves strips from Gui Re-4. The optimum callus differentiation medium were modified B5 medium supplemented with 0.75 mg/L TDZ for callus from Gui Re-6 and 0.75~1.00 mg/L TDZ for callus from Gui Re-4. To achieve 50% callus browning rate during callus proliferation, the concentration of glyphosate in medium should be 800 mg/L for Gui Re-6,and 600 mg/L for Gui Re-4. To get 50%callus browning rate during callus differentiation, the concentration of glyphosate in medium should be 600 mg/L for Gui Re-6,and 400 mg/L for Gui Re-4. The resistance test by spray of glyphosate showed that the resistant seedlings possessed stronger resistance to glyphosate than the control. Gui Re-6 resistance somaclonal variation to glyphosate was screened by two step method in callus proliferation medium and cultivation of callus differentiation. By this experiment,the screening system of resistance somaclonal variation to glyphosate was established in vitro in cassava, and this methods can be used for clone breeding studies.
