摘要
为了建立东北矮紫杉体细胞胚的诱导培养体系,以东北矮紫杉种胚为实验材料,研究了基本培养基及2,4-D、NAA、Kinetin(KT)和Zeatin(ZT)等植物生长调节剂对体细胞胚诱导效果的影响,筛选出适合诱导愈伤组织和体细胞胚的培养基配方。结果表明,种胚在mB5+0.5 mg/L 2,4-D+4 mg/LNAA+0.2 mg/L KT培养基上暗培养27 d后成功诱导出大量的愈伤组织,并且生长迅速,诱导率达66.7%;将诱导出的愈伤组织转移到mB5+3 mg/L ZT培养基中培养,20 d后表面上出现白色的颗粒状培养物,经显微观察发现白色颗粒状培养物为胚性组织,可以观察到球状细胞团。适合东北矮紫杉成熟种子诱导愈伤组织的培养基是mB5+0.5 mg/L 2,4-D+4 mg/L NAA+0.2 mg/L KT,适合诱导体细胞胚的培养基是mB5+3 mg/L ZT。
To establish the induction and culture system for the somatic embryo of Taxus cuspidate "Nana",the mature embryo of Taxus cuspidate "Nana" was used to test the effects of basic medium and plant growth regulators such as 2,4-D,NAA,Kinetin(KT) and Zeatin(ZT) on the induction of somatic embryo.The results showed that sufficient callus was induced successfully on mB5+0.5 mg/L 2,4-D+4 mg/L NAA+0.2 mg/L KT under dark after 27 d of culture,and the induction rate reached about 66.7%.In addition,the primary callus was transferred to the medium of mB5+3 mg/L ZT,and it was found that white granular culture occurred on the surface of the callus,and it was further confirmed that this culture was characteristic of embryonic tissue under light microscope.The suitable medium for the induction of callus and somatic embryo from the seeds of Taxus cuspidate "Nana" were mB5+0.5 mg/L2,4-D+4 mg/L NAA+0.2 mg/L KT and mB5+3 mg/L ZT,respectively.These results provided theoretical base for the production of high-quality seedlings through artificial seeds using somatic embryo as propagules.
