摘要
为了建立一套便捷、准确、稳定的玉米种子纯度鉴定方法,本研究以南校969玉米杂种一代及其亲本为材料,对20对均匀分布于玉米染色体上的SSR引物进行筛选,SSR引物phi085适合该品种纯度鉴定。对利用幼嫩胚芽和干种子为材料提取DNA的实验效果进行比较,建立了一套利用SSR标记技术简便高效进行玉米杂种一代纯度鉴定的技术体系。研究结果证明,利用玉米幼嫩胚芽和干种子胚提取DNA,通过一对SSR引物扩增和变性聚丙烯酰胺凝胶电泳检测,可以方便地进行玉米杂交种纯度鉴定。
In order to construct a convenient, accurate and stable seed purity identification system, In this research, we used Nanxiao 969 maize hybrid and their parents as materials, 20 SSR primer pairs were selected from maize chromosomes, and then a polymorphic SSR marker phi085 was founded that could be applied to test the purity of this variety. We compared the two methods between extracting DNA from tender germ and dried seed of maize and constructed a convenient, accurate and stable maize hybrid purity identification system by using SSR marker. And results showed that extracting DNA from tender germ or dried seed of maize and through a pair of SSR primer amplification and denaturing polyacrylamide gel electrophoresis detection can be easily to identified maize hybrid purity.
