摘要
目的:研究三七皂苷Fc对2型糖尿病db/db小鼠肾脏的保护作用。方法:8周龄雄性db/db及其同窝对照db/m小鼠分为db/m对照组,db/db模型组及db/db模型+三七皂苷Fc治疗组,每组5只。三七皂苷Fc组给予三七皂苷Fc 5 mg/(kg·d)橄榄油溶液灌胃,db/m对照组,db/db模型组给予等量橄榄油灌胃,连续8周。灌胃8周末测定小鼠体质量、血糖,收集24 h尿液,并留取血液及肾脏组织,检测尿白蛋白浓度、血肌酐及肾脏病理损伤,并采用免疫组织化学方法检测肾组织整合素连接酶(Integrin Linked Kinase,ILK)表达。结果:与db/db模型组比较,三七皂苷Fc组尿白蛋白明显降低(P<0.05),HE染色显示三七皂苷Fc可明显改善肾脏病理损伤。此外,三七皂苷Fc还可明显下调db/db小鼠肾组织ILK表达。结论:三七皂苷Fc可降低2型糖尿病db/db小鼠尿蛋白,延缓肾脏病变进展,具有肾脏保护作用。
Objective:To study the protective effects of Notoginsenoside Fc on the kidney of db/db mice. Methods: Eight weeks old male db/db and littermate control db/mice were divided into three groups:db/m group, db/db model group and Notoginsenoside FC group, 5 mice in each group. Notoginsenoside Fc group was given Notoginsenoside Fc dissolved by olive oil 5 rag/( kg · d). Db/m group and db/db model group were given equal amount of olive oil. All mice were garaged for 8 weeks. At the end of the study, the weight, blood glucose and 24 h urine volume were detected. After the experiment, the blood and kidney were collected and the urine albumin concentration, serum creatinine and renal his- topathology were detected. Results:Compared with the db/db model group, the urinary albumin in the Notoginsenoside Fe group was significantly decreased ( P 〈 0. 05 ) ; the renal histopathology were obviously improved and the expression of ILK was down regulated. Conclusion:Notoginsenoside Fc has a protective effect on the kidney of db/db mice, which can delay the progression of diabetic nephropathy.
