摘要
目的:建立以高效液相色谱法测定维参锌胶囊中人参皂苷含量的方法。方法:色谱柱为HypersilBDS-C18(125mm×4.6mm,5μm),流动相为乙腈-水,梯度洗脱(0~17min,乙腈19%,流速为1mL.min-1;17~20min,乙腈19%~31%,流速为1mL.min-1;20~35min,乙腈31%~40%,流速为1mL.min-1;35~40min,乙腈97%,流速为1.5mL.min-1;40~45min,乙腈19%,流速为1mL.min-1),检测波长为203nm,柱温为30℃。结果:人参皂苷Rg1检测浓度在0.0125~0.2000mg.mL-1(r=0.9997)、人参皂苷Re在0.01~0.16mg.mL-1(r=0.9999)、人参皂苷Rb1在0.0125~0.2000mg.mL-1(r=0.9994)范围内与峰面积积分值呈良好线性关系;平均回收率分别为93.8%、96.7%、95.3%,RSD分别为1.9%、1.6%、1.8%。结论:本方法简便、快速、准确、灵敏度高、重现性好,可用于维参锌胶囊的质量控制。
OBJECTIVE: To establish a HPLC method for the determination of Ginsenosides (Rg1, Re and Rb1) in Zinc sulfate capsule. METHDOS: Hypersil BDS- C18 (125mm × 4.6mm, 5μm) was taken as chromatographic column, and the mobile phase was composed of acetonitrile and water (0--17min, acetonitrile 19%, v(flow rate) = 1mL·min^-1; 17--20min, acetonitrile 19% --31%, v = 1mL·min^-1 20-35min, acetonitrile 31% --40%, v = 1mL·min^-1; 35--40min, acetonitrile 97%, v = 1.5mL·min^-1; 40--45min, acetonitrile 19%, v =1mL·min^-1) in a gradient elution. The detection wavelength was 203nm and the column temperature was 30℃ . RESULTS: The linear ranges of Rg1,Re and Rb1 were 0.012 5--0.2.00 0mL·min^-1(r = 0.999 7), 0.01-0.16mL·min^-1 (r =0.999 9) and 0.012 5-0.200 0mL·min^-1(r =0.999 4) respectively. The average recoveries were 93.8%, 96.7% and 95.3% respectively with RSD at 1.9%, 1.6% and 1.8% respectively. CONCLUSION: The method is simple, rapid ,accurate, sensitive and reproducible,and suitable for the quality control of Zinc sulfate capsule.
