摘要
研究了桃花总黄酮的纯化工艺及抗氧化活性。采用静态吸附-解吸和动态吸附-解吸的方法,以吸附率和解吸率为指标,从8种树脂中筛选出最佳树脂为DM-28;以总黄酮浓度为指标,优选出大孔树脂DM-28纯化桃花总黄酮的最佳工艺参数为:静态吸附:最佳温度为30℃,最适p H为4.0,吸附3.5 h,60%乙醇洗脱;动态吸附:上样浓度为0.72 mg/m L、上样流速为2 m L/min,60%乙醇以2 m L/min流速进行洗脱,在此优化条件下,纯化后的桃花总黄酮(58.39%)比纯化前提高6.84倍。此外,通过测定桃花总黄酮清除自由基的能力对其抗氧化活性进行了评价,结果表明:桃花总黄酮的抗氧化能力较强,对DPPH、ABTS+自由基的半抑制浓度(IC50)分别为13.47μg/m L、32.16μg/m L。
The conditions for purification of total flavonoids from Prunus persica by macroporous resin were explored.With adsorption-desorption ratios as indexes,optimum resin DM-28 was selected by static adsorption-desorption and dynamic adsorption-desorption ratio tests.The concentration of eluting agent was determined with the content of total flavonoids as the index. The optimum conditions for static absorption was as follows: the best temperature was 30 ℃,the value of p H 4.0,the time of absorption 3.5 h; the optimum condition for dynamic adsorption was as follows: the flow rate 2 m L / min,sample concentration 0. 72 mg / m L,and60% alcohol as eluant for desorption with the flow rate 2 m L / min. Under these conditions,the total flavonoid from Prunus persica increased by 6.84 times.Moreover,the antioxidant activity of flavonoids from Prunus persica was explored by using free radical scavenging reactions.The results indicated that Prunus persica flavonoid has an obvious antioxidant activity,the IC50 value of DPPH and ABTS+ were 13.47μg / m L,32.16 μg / m L,respectively.
