摘要
目的了解孕鼠在妊娠期糖尿病(GDM)环境下胎盘脂肪酸的转运功能,探讨维生素E(VE)干预对GDM孕鼠胎盘脂肪酸转运的影响。方法 C57BL/6J小鼠120只,试验前动物自由进食标准饲料,在光照/黑暗12h:12h、室温(20~25)℃,湿度(50±5)%环境适应1w后,按2:1雌雄合笼,次日晨7:00查阴栓,以查到阴栓为交配成功,并定为孕0日(GD0)。对孕鼠使用链脲霉素(STZ)构建GDM模型成功。选取GDM模型小鼠24只,随机均分成GDM+VE和GDM两组;另选12只孕鼠对照组(CN)。GDM+VE组,给予VE,5 mg/kg·d p.o.,CN和GDM组给予等量溶剂。GD18处死孕鼠,取血和胎盘按ELISA法、实时定量PCR法、蛋白免疫印迹法进行检测。结果与对照(CN)组比较,GDM组母鼠血清中的IL-6、IL-8明显上升(P<0.01),而TNF-α水平差异无统计学意义(P>0.05);VE干预后,与GDM组相比,VE干预组的母鼠血清中的TNF-α、IL-6、IL-8下降(P<0.05)。与CN组比较,GDM组PPARγ、FATP-1、FATP-4蛋白表达水平下调(P<0.05);与GDM组相比,VE干预组PPARγ、FATP-1、FATP-4蛋白表达水平上升(P<0.05)。另外,GDM组PPARγ、FATP-1、FATP-4的m RNA的表达水平与CN组比明显下调(P<0.01);与GDM组比,VE干预组PPARγ、FATP-1的m RNA表达水平上升(P<0.05),FATP-4的m RNA表达水平无明显变化(P>0.05)。结论 VE可抑制TNF-α、IL-6/8等炎症因子的分泌,上调PPARγ、FATP-1、FATP-4的表达。
Objective To observe the expression of placental fatty acid transporters in gestational diabetes mellitus (GDM) mice and examine the effects of vitamin E (VE).Methods A total of 120 C57BL/6J mice were included in this study. They were allowed to free access to food and water at all times and maintained on a 12-h light/dark cycle in the controlled temperature (20-25°C) and humidity (50±5) % environment for one week. Subsequently, male and female mice were housed together at a 1: 2 ratio. In the following morning, mating was marked by the presence of a vaginal mucous plug which was defined gestation day (GD) 0. Pregnant mice were induced gestational diabetes mellitus through intraperitoneal administration of streptozotocin (STZ). The 36 pregnant mice was randomly assigned into three groups: CN (the control group), GDM, GDM+VE (the study group). Each group had 12 mice. The mice in the GDM+VE group were administrated with 5 mg/kg VE. The mice in CN group and GDM group were given the same amount of solvent. At GD18, studied mice were euthanized by intraperitoneal injection of sodium pentobarbital and their placentas were collected immediately then detected by ELISA, real-time PCR and Western-blot.Results Compared with the CN mice, GDM mice had a significant high level of serum IL-6, IL-8 (P〈0.01), but TNF-α level was without significant difference (P〉0.05). TNF-α, IL-6, IL-8 were significantly lower in mice after the VE intervention (P〈0.05). The PPARγ, FATP-1, FATP-4 protein levels in GDM mice placenta were lower than these in CN mice (P〈0.05). However, the PPARγ, FATP-1, FATP-4 levels were significantly higher in mice after the VE intervention (P〈0.05). The expressions of PPARγ, FATP-1, FATP-4 mRNA levels were reduced significantly in GDM mice placenta, compared with these in CN mice placenta(P〈0.01); and the levels of PPARγ and FATP-1 proteins were higher compared with that in GDM mice significantly(P〈0.05), while FATP-4 mRNA
