摘要
以转2mG2-epsps基因抗草甘膦水稻为材料,根据实质等同性原则,分析转基因(genetically modified,GM)水稻种子可溶性蛋白含量和组分的非预期变异。结果表明:与非转基因亲本相比,GM水稻种子的总可溶性蛋白含量极显著降低,主要受谷蛋白含量变异影响。但清、球、醇、谷4种蛋白含量的最高变异幅度(9.12%)低于P1和P2两亲本之间的最大差异幅度(14.67%)。说明GM水稻种子的可溶性蛋白含量非预期变异在安全范围内,但营养品质可能与亲本品系存在差异。SDS-PAGE分析发现,GM水稻株系T12和T22分别有12和14个蛋白谱带发生了非预期变异,涉及4种可溶性蛋白的组分,多数表现为含量变化。突出的是,2个GM水稻株系中均新增加了41 ku和56 ku谷蛋白组分,需要进一步通过毒理学分析评价其食品安全性。
The unintended variation of soluble protein contents and components of genetically modified (GM) rice seeds were analyzed, using glyphosate - resistant GM rice lines harboring 2raG2 - epsps gene as materials, ac- cording to the principle of substantial equivalence. The results showed that the total soluble protein contents of GM rice seeds decreased very significantly, which were mainly affected by the glutenin content variation, compared to the non -transgenic parent lines. While the maximum content variation range (9.12%) from four kinds of proteins in- cluding albumin, globulin, prolamin, glutenin, was lower than the variation difference (14.67%) between the two parent varieties, P1 and P2. This indicated that the unintended variation of soluble protein contents of GM rice lines was within the safety range, but the nutritional quality of GM rice seeds might be different from their parent varieties. SDS -PAGE analysis reveals that there were 12 and 14 unintended variation protein bands in the respective GM rice line T12 and T22, concerning components of four kinds of soluble proteins and most exhibit content changes. Nota- bly, two new glutenin bands of 41 ku and 56 ku were produced in all the two GM rice lines, which must be to further toxicology test for food safety evaluation. subjected
