摘要
目的:寻求能有效防治糖尿病神经病变的最佳中药复方制剂。方法:采用L18(37)正交表,考查三因素(葛根提取物、黄芪提取物和丹参提取物)在三水平(20mg/L,100mg/L,200mg/L)上预处理对低浓度(5mmol/L)过氧化氢(H_2O_2)致大鼠脑皮质线粒体损伤SOD酶活性的保护作用;考查三因素(葛根提取物、黄芪提取物和丹参提取物)在三水平(20mg/L,40mg/L,100mg/L)上预处理对高浓度(300mmol/L)H_2O_2致大鼠皮质脑线粒体损伤Na^+,K^+-ATP酶活性的保护作用。结果:经5mmol/L或300mmol/LH_2O_2处理的线粒体,其SOD活力以及Na^+,K^+-ATP酶活性均显著降低,与对照组比较有统计学差异。线粒体SOD活力分析结果显示:黄芪提取物和丹参提取物在增加SOD活性方面发挥主要作用,葛根*黄芪、葛根*丹参以及葛根*黄芪*丹参之间存在显著的协同增强作用。线粒体Na^+,K^+-ATP酶活性分析结果显示:黄芪提取物和丹参提取物在增加Na^+,K^+-ATP酶活性方面发挥主要作用,并且葛根*黄芪*丹参之间存在显著的协同增强作用。结论:由葛根提取物,黄芪提取物和丹参提取物以1:1:1剂量比例组成的中药复方制剂对H_2O_2所致大鼠脑线粒体损伤具有最佳保护作用。
Objective: To seek the best prescription of Chinese medicine compound to prevent and cure diabetic neuropathy. Methods: Doing just as L18( 37) orthogonal table,the mitochondria from the cortex of SD rat brains were pretreated with the three factors( the extracts of lobed kudzuvine root,the extracts of astragalus membranaceus( Fisch.) Bunge,the extracts of salvia miltiorrhiza Bge) and three levels( 20 mg/L,100 mg/L,200 mg/L) and 5mmol/L hydrogen peroxide( H_2O_2) or with the three factors( the extracts of lobed kudzuvine root,the extracts of astragalus membranaceus( Fisch.) Bunge,the extracts of salvia miltiorrhiza Bge) and three levels( 20 mg/L,40 mg/L,100 mg/L)and 300 mmol/L H_2O_2,the the activity of SOD or the Na^+,K^+-ATP were measured. Results: The mitochondria were treated by the H_2O_2 of 5mmol/L or 300 mmol/L,the activities of SOD and Na^+,K^+-ATP were decreased significantly. The activity of mitochondria SOD analysis showed that: the extracts of astragalus membranaceus( Fisch.) Bunge and the extracts of salvia miltiorrhiza Bge played a major role,there were synergistic effects between the extracts of lobed kudzuvine root and the extracts of astragalus membranaceus( Fisch.) Bunge,as well as the extracts of lobed kudzuvine root and the extracts of salvia miltiorrhiza Bge,aslo among the extracts of lobed kudzuvine root,the extracts of Astragalus membranaceus( Fisch.) Bunge and the extracts of salvia miltiorrhiza Bge in increasing the activity of SOD. The activity of mitochondrial Na^+,K^+-ATP analysis showed that: the extracts of astragalus membranaceus( Fisch.) Bunge and the extracts of salvia miltiorrhiza Bge played major roles. There were synergistic effects among the extracts of lobed kudzuvine root,the extracts of astragalus membranaceus( Fisch.) Bunge and the extracts of salvia miltiorrhiza Bge in increasing the activity of Na^+,K^+-ATP. Conclusion: the Chinese medicinal prescription which the dose composition ratio of the
