摘要
中医认为金钗石斛(Dendrobium nobile)、麦冬(Ophiopogon japonicas)兼具有养阴生津的作用,能滋阴润燥。为了验证它们在皮肤保湿方面的功效,发掘更多可应用于化妆品的天然活性物,开展试验研究。将体外培养永生化人皮肤角质形成细胞(Ha Ca T)给予金钗石斛、麦冬以及不同比例复配的金钗石斛麦冬提取物处理,通过MTT比色法和形态学观察,检测处理对表皮细胞活性影响;荧光定量PCR(realtime fluores-cence quantitative PCR)检测与皮肤保湿相关的水通道蛋白3(aquaporin3,AQP3)、丝聚蛋白(filaggrin,FLG)和半胱氨酸天冬氨酸特异性蛋白酶-14(caspase-14,CAS14)三个基因的m RNA表达水平。试验结果表明,金钗石斛、麦冬单药具有提高AQP3的m RNA表达水平的作用,两者的复配提取物,在质量比1︰1和3︰4下,分别对这三个基因中的一个或两个基因的m RNA表达水平有提升作用,而在质量比4︰3时,则对AQP3、FLG和CAS14三个基因的m RNA表达水平均有显著提升作用。金钗石斛、麦冬以及其复配提取物具有促进保湿相关基因表达的作用,提示它们在皮肤保湿方面具有应用价值。
According to traditional Chinese medicine theory,both Dendrobium nobile and Ophiopogon japonicus have the function of nourishing yin and promoting body fluid,and moistening dryness.In order to verify their efficacy in skin moisturizing,explore more natural active ingredients that can be used in cosmetics,experimental research was carried out.Immortalized human keratinocytes(HaCaT)were cultured in vitro,treated with Dendrobium nobile,Ophiopogon japonicus,and different mass proportion of compound extracts.The effects on the activity of HacaT cells were detected by MTT colorimetry and morphological observation.The mRNA expression levels of aquaporin 3(AQP3),fibroin(FLG)and caspase-14(CAS14)were detected by real-time fluorescence quantitative PCR.The experimental results show that the single use of Dendrobium nobile or Ophiopogon japonicus can improve the expression level of AQP3 mRNA,and the mixture of these two extracts,at mass ratio of 1/1 and 3/4,can enhance the mRNA expression level of one or two genes,respectively,while at mass ratio of 4/3,it can significantly promote the mRNA expression level of AQP3,FLG and CAS14.Dendrobium nobile,Ophiopogon japonicus and their compound extracts can affect the expression of skin moisturizing related genes,suggesting their application value in skin moisturizing.
