摘要
目的探讨同源转录因子2(CDX2)、环氧合酶-2(COX-2)、核因子-κB(NF-κB)对胃癌患者病情进展的预测价值及其相互关系。方法应用免疫组织化学法和反转录聚合酶链反应检测30例正常胃黏膜组织、42例胃癌癌前病变组织及34例胃癌组织中CDX2、COX-2、NF-κB蛋白及其m RNA的表达水平。结果胃癌组织和癌前病变组织中CDX2、COX-2、NF-κB m RNA的表达显著高于正常胃黏膜组织(P<0.05),胃癌组织中COX-2、NF-κB m RNA的表达显著高于癌前病变组织(P<0.05),胃癌组织中CDX2 m RNA的表达显著低于癌前病变组织(P<0.05)。胃癌组织和癌前病变组织中CDX2、COX-2、NF-κB蛋白阳性表达率显著高于正常胃黏膜组织(P<0.05),胃癌组织中COX-2、NF-κB蛋白阳性表达率显著高于癌前病变组织(P<0.05),胃癌组织中CDX2蛋白阳性表达率显著低于癌前病变组织(P<0.05)。CDX2表达与胃癌临床分期、分化程度有关(P<0.05),COX-2和NF-κB表达与临床分期、分化程度、肿瘤直径及淋巴结转移有关(P<0.05)。CDX2与COX-2、NF-κB表达呈负相关(P<0.05),COX-2与NF-κB表达呈正相关(P<0.05)。结论 CDX2表达下调及COX-2、NF-κB表达上调可能与胃癌的发生发展、浸润及淋巴结转移有关,通过对三者表达及关系进行分析有助于了解胃癌患者的病情进展及预后。
Objective To investigate the value of homologous transcription factor 2 (CDX2) ,cyclooxygenase-2 ( COX- 2) and nuclear factor-κB (NF-κB) in predicting the progression of gastric cancer and their relationships. Methods The expressions of CDX2,COX-2,NF-κB protein and mRNA in 30 cases of normal gastric mucosa tissue,34 cases of precancerous tissue and 42 cases of gastric cancer tissue were detected by immunohistoehemical method and reverse transeription-polymerase chain reaction. Results The expressions of CDX2, COX-2 and NF-κB mRNA in gastric cancer tissue and precancerous tissue were significantly higher than those in normal gastric mucosa tissue( P 〈 0.05 ). The expressions of COX-2 and NF-κB mRNA in gastric cancer tissue were significantly higher than those in precancerous tissue (P 〈 0. 05 ). The expression of CDX2 mRNA in gastric cancer tissue was significantly lower than that in precancerous tissue ( P 〈 0.05 ). The expressions of CDX2, COX-2 and NF-KB protein in gastric cancer tissue and precancerous tissue were significantly higher than those in normal gastric mueosa tissue (P 〈 0.05 ). The expressions of COX-2 and NF-κB protein in gastric cancer tissue were significantly higher than those in precancerous tissue( P 〈 0.05 ). The expression of CDX2 protein in gastric cancer tissue was significantly lower than that in precancerous tissue ( P 〈0O. 05 ). CDX2 expression was related to clinical stage and differentiation degree of gastric cancer ( P 〈 0.05 ). The expression of COX-2 and NF-κB was related to clinical stage, differentiation degree, tumor diameter and lymph node metasta- sis ( P 〈 0.05 ). CDX2 expression was negatively correlated with COX-2 and NF-κB ( P 〈 0.05 ). COX-2 expression was positively correlated with NF-κB (P 〈 0.05). Conclusions The down-regnlation of CDX2 expression and the up-regnlation of COX-2 and NF-κB expression may be related to the occurrence, development,invasion and lymph node metastasis of gastri
