摘要
目的观察不同中药复方对心肌梗死后心力衰竭大鼠心肌组织中Ⅰ、Ⅲ型胶原的影响。方法 2010年4月—2011年1月,以冠状动脉结扎配合力竭式游泳的方法造成心肌梗死后心力衰竭大鼠模型,分为模型组、西药组、益气组、活血组、益气活血组,另设空白对照组。每组10只大鼠。采用实时荧光定量PCR及免疫组化检测心肌梗死后心力衰竭大鼠心肌组织中Ⅰ、Ⅲ型胶原的表达。结果实时荧光定量PCR和免疫组化结果均显示:与空白对照组比较,模型组大鼠Ⅰ、Ⅲ型胶原表达下降(P<0.05);与模型组比较,各给药组大鼠Ⅰ、Ⅲ型胶原表达升高(P<0.05);西药组、益气活血组大鼠Ⅰ、Ⅲ型胶原表达高于益气组及活血组(P<0.05)。结论益气活血中药复方较益气中药复方、活血中药复方更能提高心肌梗死后心力衰竭大鼠心肌组织中Ⅰ、Ⅲ型胶原的表达,可达到治疗心力衰竭的作用。
Objective To observe the effect of different Chinese herbal compound on type Ⅰ,Ⅲ collagen in myocardium of rats with heart failure following myocardial infarction. Methods From April 2010 to January 2011,the rat model of heart failure following myocardial infarction were established by coronary artery ligation method and exhaustive swimming. The experimental rats were divided into model group, western medicine group, Reinforcing qi group, Activating blood group,Reinforcing qi and activating blood group,and control group. 10 rats in each group. Real time fluorescence quantitative PCR and immunohistochemistry were used to detect the expression of type Ⅰ,Ⅲ collagen in the myocardium of rats with heart failure following myocardial infarction. Results Compared with control group,the expression of type Ⅰ,Ⅲ collagen in model group detected by PCR and immunohistochemistry significantly decreased( P〈 0. 05). Compared with model group,the expression of type Ⅰ,Ⅲ collagen in each treatment groups detected by PCR and immunohistochemistry significantly increased( P 〈0. 05).The expression of type Ⅰ,Ⅲ collagen in western medicine group and Reinforcing qi and Activating blood group detected by PCR and immunohistochemistry were significantly higher than Reinforcing qi group and Activating blood group( P〈 0. 05).Conclusion Reinforcing qi and activating blood Chinese herbal compound can improve the expression of type Ⅰ,Ⅲ collagen in myocardium of rats with heart failure following myocardial infarction,thus achieve the role of treatment of heart failure.
