摘要
目的:研究三七总皂苷对血管紧张素Ⅱ(AngⅡ)诱导培养心肌细胞凋亡的影响。方法:原代培养乳鼠心肌细胞,通过MTT法观察三七总皂苷对培养心肌细胞活力的影响;AO荧光染色观察细胞核形态改变,流式细胞术检测细胞凋亡,Fluo3荧光探针标记测定钙离子荧光强度。结果:AngⅡ(10-7mol·L-1)孵育48h后心肌细胞凋亡明显多于对照组;三七总皂苷(25,50,100mg·L-1)组心肌细胞活力显著高于AngⅡ组;三七总皂苷(50mg·L-1)组AngⅡ诱导的心肌细胞凋亡率明显低于AngⅡ组;三七总皂苷(50mg·L-1)组心肌细胞内钙超载明显轻于AngⅡ组。结论:三七总皂苷对AngⅡ诱导的细胞凋亡具有明显的抑制作用,其机制可能与抑制心肌细胞内钙超载有关。
Objective: To study the protective effects of Panax notoginseng saponins(PNS) on angiotensinⅡ(AngⅡ)-induced rat cardiomyocyte apoptosis in vitro and the probable mechanism. Method: Cultured cardiomyocytes from neonatal rats were stimulated with AngⅡ. Cell viability was measured by MTT. Apoptosis was evaluated using Acridine Orange(AO) fluorescent dye staining and flow cytometry; Fluo-3 AM was used to test the change of intracellular free calcium. Result: It was found that incubating with AngⅡ(10^(-7) mol·L^(-1)) for 48 h increased cardiomyocyte apoptosis, PNS (25,100 mg·mL^(-1)) increased myocyte viability. PNS(50 mg·mL^(-1)) significantly decreased this AngⅡ-induced rat cardiomyocyte apoptosis(P<0.05) and decreased fluorescent intensity of intracellular calcium.Conclusion: PNS has a significant effect on AngⅡ-induced rat cardiomyocytes apoptosis in vitro by alleviating intracellular calcium overload.
