摘要
目的探讨壮筋续骨汤对股骨骨折大鼠骨痂组织中Ⅰ型胶原表达的影响。方法健康雄性Wistar大鼠50只,随机分为假手术组、模型组、对照组和治疗组。模型组、对照组和治疗组大鼠用线锯切断股骨中段制备股骨骨折模型。治疗组大鼠给予壮筋续骨汤灌胃每天1次,连续28d,其他组给予等量生理盐水灌胃。对照组和治疗组大鼠分别于骨折7、14、21、28d时取股骨标本观察骨折局部愈合情况,苏木精-伊红染色观察骨折的病理变化,免疫组织化学染色检测骨痂组织中Ⅰ型胶原的表达。结果假手术组大鼠骨质形态正常,Ⅰ型胶原微弱表达;模型组大鼠骨折当天骨折断端被血肿组织填充,Ⅰ型胶原表达显著增强。对照组和治疗组大鼠骨折7、14、21、28d时骨折断端分别被肉芽组织、纤维性骨痂、软骨性骨痂、骨性骨痂填充,但治疗组大鼠骨折28d时骨痂组织结构明显优于对照组。骨折后7、14、21、28d,对照组和治疗组大鼠骨痂组织中Ⅰ型胶原表达水平逐渐减弱,但治疗组大鼠骨折7、14d时Ⅰ型胶原表达水平显著高于对照组(t=2.83、3.42,P<0.05)。结论壮筋续骨汤在骨折早期能促进骨痂组织中Ⅰ型胶原的合成,有利于纤维-软骨性骨痂的形成。
Objective To explore the effect of Zhuangjin Xugu（ZJXG）decoction on the expression of typeⅠcollagen in callus tissue of femur fracture rats. Methods Fifty healthy adult male Wistar rats were randomly divided into sham-operation group,model group,control group and treatment group.A femur-fracture model was created in model group,control group and treatment group by cutting the middle femur with a wire saw.The rats in treatmet group were treated with intragastric administration of ZJXG decoction,once a day,for 28 days.Those in the other groups were treated with intragastric administration of equivalent normal saline.Femoral specimens of rats in the control and treatment groups were taken on days 7,14,21 and 28after fracture to inspect its local healing.The pathological changes of the fracture were investigated with hematoxtlin-eosin（HE）staining.The expression of type Ⅰ collagen in callus was evaluated using immunohistochemical staining. Results In the sham operation group,the shape and structure of femur were normal with weak expressions of typeⅠcollagen.In model group,on fracture day,the broken ends of the fracture were filled with hematoma,the expressions of typeⅠcollagen increased markedly.In control group and treatment group,on days 7,14,21 and 28of fracture,the broken ends of the fracture were filled with granular tissue,fibrous callus,cartilaginous callus and osseous callus,respectively,and in the treatment group,the callus structure on day 28 was markedly better than that in the control group.After 7,14,21 and 28days of fracture,the expression of typeⅠ collagen in the control group and treatment group gradually decreased,but on days 7and 14,the expression of typeⅠ collagen in the treatment group was much higher than that in the control group（t=2.83,3.42;P〈0.05）. Conclusion ZJXG decoction can boost the synthesis of typeⅠ collagen in callus in early stage of fracture,which is conducive to the formation of fibrous-cartilage callus.
