摘要
目的:建立同时测定玉屏风口服液中1个异黄酮类成分(毛蕊异黄酮葡萄糖苷)及3个色原酮类成分(升麻素苷、升麻素、5-O-甲基维斯阿米醇苷)含量的高效液相色谱法。方法:采用Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-水(17∶83)为流动相,流速1.0 m L·min^(-1),检测波长为254 nm。结果:毛蕊异黄酮葡萄糖苷、升麻素苷、升麻素和5-O-甲基维斯阿米醇苷进样量分别为0.127~2.536μg(r=0.999 9)、0.097~1.944μg(r=0.999 9)、0.081~1.616μg(r=0.999 9)和0.202~4.040μg(r=0.999 9)范围内与峰面积呈现良好的线性关系;平均回收率(n=6)分别为99.8%、99.2%、99.9%和100.5%,RSD分别为1.0%、1.2%、1.0%和1.0%。测得毛蕊异黄酮葡萄糖苷、升麻素苷、升麻素和5-O-甲基维斯阿米醇苷的含量分别为0.100~0.101、0.087~0.089、0.074~0.075和0.183~0.185 mg·m L^(-1)。结论:经方法学验证,本文建立的方法可用于玉屏风口服液的质量控制。
Objective： To develop an HPLC method for simultaneous determination of one isoflavonoid （calycosin-7-O-β-D-glucoside） and three chromones （prim-O-glucosylcimifugin, cimifugin, 4＇-O-β-glucopyranosyl-5-O-methylvisamminol） in Yupingfeng oral liquid.Methods： The separation was carried out on a Diamonsil C18 （250 mm×4.6 mm, 5 μm） with the mobile phase of acetonitrile-water （17：83）.The flow rate was 1.0 mL·min-1, and the detection wavelength was 254 nm Results： The linear range of calycosin-7-O-β-D-glucoside, prim-O-glucosylcimifugin, cimifugin and 4＇-O-β-glucopyranosyl-5-O-methylvisamminol were 0.127-2.536 μg （r=0.999 9）, 0.097-1.944 μg （r=0.999 9）, 0.081-1.616 μg （r=0.999 9） and 0.202-4.040 μg （r=0.999 9） within the scope of good linear with peak area present relationship.The average recoveries （n=6） were 99.8% （RSD=1.0%）, 99.2% （RSD=1.2%）, 99.9% （RSD=1.0%）, 100.5% （RSD=1.0%）, respectively; the contents of the above-mentioned compounds in samples were 0.100-0.101 mg·mL-1, 0.087-0.089 mg·mL-1, 0.074-0.075 mg·mL-1 and 0.183-0.185 mg·mL-1, respectively.Conclusion： The established method is proved by methodology validation that it can be used in the quality control of the Yupingfeng oral liquid.
